The speed at which most countries with high burdens of multidrug-resistant tuberculosis (MDRTB) have scaled up their capacity to diagnose and treat individuals with these forms of TB has failed to keep pace with the problem. Limited availability of drug susceptibility testing, high costs and inefficiencies in the supply of second-line drugs, and inadequate capacity for the management of patient...
Currently, no agar-based susceptibility testing method has been standardized for testing dermatophytes. We describe a newly developed agar-based method employing disk diffusion assay to test the susceptibility of 47 isolates of dermatophytes against 8 antifungals. Our results show that the method is reproducible, is simple, and could be used to determine the antifungal susceptibility of dermato...
The critical concentration (CC) for ethambutol testing on the Bactec MGIT 960 M. tuberculosis susceptibility testing has been questioned in recent publications. In this study, we correlate susceptibility results from the Bactec 460, MGIT 960 and embB gene sequencing to determine if the Bactec MGIT 960 adequately detects ethambutol resistance. We discovered discrepancies between the methods that...
Background and Objectives:Infection of pleural fluid is a common disease and because of antibiotic administration, the microbiology of this fluid has changed. The aim of this study was to determine the common bacteria and suitable antibiotics for treatment in pleural effusion (PE). Materials and Methods: In this cross sectional study, 1210 samples with exudative features were cultured for p...
This short paper presents a preliminary description of two new protocols for privacy-preserving disease susceptibility testing, following the model proposed by Ayday et al. in [4]. We show that an alternative encoding of the patient’s SNPs can simplify private computations, and make patient-side computation on a trusted smartcard device extremely efficient. To support larger tests, we propose a...
One hundred clinical isolates of Candida albicans were tested for amphotericin B and fluconazole susceptibilities by the National Committee for Clinical Laboratory Standards (NCCLS) broth microdilution test at center 1 (C1). The same isolates were tested blinded at center 2 (C2) by NCCLS and flow cytometry (FC) methods. The agreement between NCCLS and FC methods ranged from 96 to 99%.
We performed Etest, disk diffusion, and broth microdilution susceptibility testing of 2,171 clinical isolates of Candida spp. against posaconazole. By using provisional breakpoints for comparison purposes only, the categorical agreement between the agar-based methods and broth microdilution results ranged from 93 to 98%, with <1% very major errors. The essential agreement (within 2 well dilutio...
Agar dilution antimicrobial susceptibility testing (CLSI, M11-A7, 2007) performed for 208 toxin-producing clinical isolates of Clostridium difficile resulted in OPT-80 MICs ranging from 0.06 to 1 microg/ml, with 90% of the isolates inhibited by a concentration of 0.5 microg/ml. The in vitro activity of OPT-80 was independent of the susceptibilities of isolates to nine other antimicrobial agents.
BACKGROUND Quality control (QC) in diagnostic microbiology is a matter of effective, efficient, accurate reporting in the expected turnaround time. Major stages of the analytical "standard operational procedures" where QC could be easily affected include organism identification and antibacterial susceptibility testing. AIM The objective of this experiential technical note is to provide an evi...
Traditional susceptibility testing of blood cultures requires overnight incubation in order to obtain isolated colonies. Susceptibility results can be reported up to 24 h sooner by using a bacterial pellet from the blood culture broth. This study evaluated the accuracy of direct susceptibility testing from positive ESP blood culture broths by using Sensititre broth microdilution plates compared...
