The hydrolysis of specifically labeled phosphatidylethanolamine and the 1 and 2 isomers of lysophosphatidylethanolamine catalyzed by enzymes in three subcellular fractions of Escherichia coli (wall, inner membrane and cytosol) has been studied. Two hydrolytic activities, phospholipases A1 and AS, are localized in the wall. Both enzymatic activities manifest similar properties and requirements f...

Plipastatins have been isolated as part of a program designed to find inhibitors of porcine pancreatic phospholipase A2. They were purified from fermentation broth of Bacillus cereus BMG302-fF67 and finally separated into four fractions by reverse phase HPLC. The respective fractions were designated as plipastatins A1, A2, B1 and B2. Plipastatins also inhibited phospholipases C and D.

Platelet activation initiates an upsurge in polyunsaturated (18:2 and 20:4) lysophosphatidic acid (LPA) production. The biochemical pathway(s) responsible for LPA production during blood clotting are not yet fully understood. Here we describe the purification of a phospholipase A(1) (PLA(1)) from thrombin-activated human platelets using sequential chromatographic steps followed by fluorophospho...

BACKGROUND Yeast surface display is a technique, where the proteins of interest are expressed as fusions with yeast surface proteins and thus remain attached to the yeast cell wall after expression. Our purpose was to study whether allergens expressed on the cell surface of baker's yeast Saccharomyces cerevisiae preserve their native allergenic properties and whether the yeast native surface gl...

The Ca(2+)- and phospholipid-binding protein Anx-A1 (annexin 1; lipocortin 1) has been described both as an inhibitor of phospholipase A(2) (PLA(2)) activity and as a mediator of glucocorticoid-regulated cell growth and eicosanoid generation. Here we show that, when compared with Anx-A1(+/+) cells, lung fibroblast cell lines derived from the Anx-A1(-/-) mouse exhibit an altered morphology chara...
