Vaccines based on recombinant proteins avoid the toxicity and antivector immunity associated with live vaccine (for example, viral) vectors, but their immunogenicity is poor, particularly for CD8(+) T-cell responses. Synthetic particles carrying antigens and adjuvant molecules have been developed to enhance subunit vaccines, but in general these materials have failed to elicit CD8(+) T-cell res...
This paper discusses the unprecedented microscopic findings of micellar growth in colloidal system (CS) of catalyzed piperidinolysis of ionized phenyl salicylate (PS-). The giant vesicles (GV) was observed under the optical polarization microscope (OPM) at [NaX]=0.1M where X=3-isopropC6H4O-. The conditions were rationalized from pseudo-first-order rate constant, kobs of PS- of micellar phase at...
The effect of 5th generation polyamidoamine (PAMAM G5) dendrimers on multilamellar dipalmitoylphosphocholine (DPPC) vesicles was investigated. PAMAM was added in two different concentrations to the lipids (10(-3) and 10(-2) dendrimer/lipid molar ratios). The thermal behavior of the evolved systems was characterized by DSC; while the structure and the morphology were investigated with small- and...
All the liposome preparation protocols, which involve drug encapsulation are multi-step processes, i.e. they consist of one or several steps of preparation and homogenization. The conditions of converting all lipids into vesicles smaller than 200 nm were determined by replacing ultrasonication with mechanical stirring of the buffer and solution of lipids in a low-boiling point organic solvent o...
Electroformation of cell-sized lipid membrane vesicles (giant vesicles, GVs), from egg yolk phosphatidylcholine, was examined varying the shape of the counter electrode. Instead of a planar ITO (indium tin oxide) electrode commonly used, platinum wire mesh was employed as a counter electrode facing lipid deposit on a planar formation electrode. The modification did not significantly alter GV fo...
The morphological changes of phosphonate polypeptoid electrolyte membranes, poly-N-(2-ethyl)hexylglycine-block-poly-N-phosphonomethylglycine (pNehm-b-pNpmn), in hydrated and dry states were characterized by cryogenic transmission electron microscopy (cryo-TEM) tomography (cryo-ET). analysis 3D tomograms revealed that the pNeh9-b-pNpm9 thin films absorbed a large amount water, resulting formatio...
Self-assembled materials such as lyotropic liquid crystals offer a wide variety of structures and applications by tuning the composition. Understanding behavior under flow induced alignment is wanted in order to tailor structure related properties. A method visualize anisotropy ordered systems situ dynamic conditions presented where flow-induced nanostructural microfluidic channels observed sca...
Supramolecular assemblies of novel aminonucleoside phospholipids and their bonding to nucleic acids.
A novel class of aminonucleoside phospholipids has been developed. These molecules could spontaneously assemble into supramolecular structures including multilamellar organization, hydrogels, superhelical strands, and vesicles. Their ability to bind to DNA by hydrogen bonding and π-π stacking interactions was investigated by many means.
