A study of the three-dimensional structure of the upper jejunal mucosa in diabetics has been carried out. The structural findings were related to 14C-L-phenylalanine uptake in vitro, sucrase activity in mucosal homogenates, and the enzyme content of the absorptive cells as measured cytophotometrically. A low grade mucosal transformation of the sprue-type was found, which was associated with dec...

Actinoplanes missouriensis utilizes arogenate as an intermediate in L-tyrosine biosynthesis, while no evidence of prephenate dehydrogenase was observed. Arogenate dehydrogenase has been partially purified by a five-step procedure. The enzyme requires NAD as cofactor. The Km values for NAD and arogenate are 0.2 mM and 0.15 mM, respectively. The molecular weight of arogenate dehydrogenase is abou...

Sodium N-[(trimethylamineboryl)-carbonyl]-L-phenylalanine 2 and {N-[(trimethylamineboryl)-carbonyl]-L-phenylalanyl- carbxylato}-bis-{N-[(trimethylaminebryl)-carbonyl]-L-phenylalanine} dicopper (II) 3 were successfully synthesized. The agents blocked L(1210) leukemic cell DNA and RNA syntheses by inhibiting multiple enzyme activities for nucleic acid synthesis, e.g. PRPP amido transferase, IMP d...

We created a novel chimeric amine dehydrogenase (AmDH) via domain shuffling of two parent AmDHs ('L- and F-AmDH'), which in turn had been generated from leucine and phenylalanine DH, respectively. Unlike the parent proteins, the chimeric AmDH ('cFL-AmDH') catalyzes the amination of acetophenone to (R)-methylbenzylamine and adamantylmethylketone to adamantylethylamine.

The prephenate dehydrogenase component of the bifunctional T-protein (chorismate mutase:prephenate dehydrogenase) has been shown to utilize L-arogenate, a common precursor of phenylalanine and tyrosine in nature, as a substrate. Partially purified T-protein from Klebsiella pneumoniae and from Escherichia coli strains K 12, B, C and W was used to demonstrate the utilization of L-arogenate as an ...

In plants, the amino acids tyrosine and phenylalanine are synthesized from arogenate by arogenate dehydrogenase and arogenate dehydratase, respectively, with the relative flux to each being tightly controlled. Here the characterization of a maize opaque endosperm mutant (mto140), which also shows retarded vegetative growth, is described The opaque phenotype co-segregates with a Mutator transpos...
