Although they decompose rapidly to produce cyanocarbenes, ethynyl azides were generated from (chloroethynyl)arenes and trapped for the first time by 1,3-dipolar cycloaddition at cyclooctyne.

The fi rst az.ide-contain ing sugar, a glycosyl azide. was reponed in 1930 by Bertho. j Since that lime various methods have been developed for the introduction of azides at d ifferent positions of sugars. A survey of available methods is given in Section 16.2. Unlillhc lale I 970s, azidcs contai ned in carbohydrate deri vatives were simply used as accessible syn-thons [or amities because of th...

Strain-promoted cycloadditions of nitrones with cyclooctynes (k(2) = 1.5 M(-1) s(-1) at 25 degrees C) are up to 25 times more rapid than comparable reactions of azides.

A one-step, large-scale preparation of alkyne-containing hyper-branched polyglycerols (HPG) is reported. The HPGs undergo click reactions to organic azides allowing a range of applications.

We report the first design of a topochemical reaction between two non-identical reactants using a supramolecular chemistry approach. A coassembly of two sugar-based organogelators with complementary reacting motifs, viz. azides and alkynes, undergoes topochemical Huisgen reaction between them.
