A single cardiomyocyte is a vital tool in the cellular and subcellular level studies of cardiac biology diseases as fundamental unit contraction electrical activity. Hence, isolating viable, high-quality cardiomyocytes from heart initial most crucial experimental step. Comparing various protocols for adult mice, Langendorff retrograde perfusion successful reproducible method reported literature...

In 1866, Carl Ludwig together with Elias Cyon created the first isolated perfused frog heart preparation. Perfusion systems for the isolated mammalian heart were developed by H. Newell Martin in 1883 and by Oscar Langendorff in 1895. In its working mode, the isolated perfused rat heart was established in the 1960s.

Objective: The goal of this study was to determine whether PDE4D regulates basal cardiac contractility. Methods and Results: At 10 to 12 weeks of age, baseline cardiac contractility in PDE4D-deficient (PDE4D / ) mice was elevated mice in vivo and in Langendorff perfused hearts, whereas isolated PDE4D / cardiomyocytes showed increased whole-cell Ca transient amplitudes and SR Ca content but unch...
