Peptide-mercaptopropionylcysteine (MPA-Cys) thioesters show a surprisingly high reactivity in native chemical ligation (NCL) and allow thiol-additive free reactions. This facilitates sequential NCL reactions and ligation-desulfurization reactions in one-pot formats. The synthetic utility is demonstrated by the synthesis of a SH3 domain.

In the presence of 2-mercaptoethanol peptide-(α)thioesters undergo smooth conversion to their corresponding peptide-(α)carboxylates. This general and operationally simple reaction extends the utility of a promising new strategy for cleaving resin-bound Boc/benzyl-protected peptides without the use of hydrogen fluoride.

Isomerically pure enolsilanes of aryl ketones, acylpyrroles, and thioesters added to the azo-imide in greater than 95% ee. The use of an alcohol additive was critical to achieving catalyst turnover.
