Enzymes, the catalysts of biological systems, are remarkable molecular devices that determine the patterns of chemical transformations. For many decades, a great deal of pure and applied studies have been devoted to enzyme reactions, as the mechanisms underlying the special characteristics of the reactions, such as high specificity and high efficiency, have not only attracted scientific interes...

Isothermal titration calorimetry is presented to be an excellent method of detecting lipid membrane solubilization upon the addition of detergents. As an example, the titration of the nan-ionic detergent C t2EOt, to the lipid POPC is studied. The main contribution to the measured titration heat is the detergent's enthalpy difference upon transfer between micelles and membranes. The detergent ad...

The purposes of this paper are (a) to examine the effect of calorimeter time constant (τ) on heat rate data from a single enzyme injection into substrate in an isothermal titration calorimeter (ITC), (b) to provide information that can be used to predict the optimum experimental conditions for determining the rate constant (k2), Michaelis constant (KM), and enthalpy change of the reaction (ΔRH)...

Isothermal titration calorimetry (ITC) has been applied to the study of proteins for many years. Its use in the biophysical analysis of RNAs has lagged significantly behind its use in protein biochemistry, however, in part because of the relatively large samples required. As the instrumentation has become more sensitive, the ability to obtain high quality data on RNA folding and RNA ligand inte...

Thermodynamics of the binding of antibiotic streptomycin to bovine serum albumin have been studied using isothermal titration calorimetry in combination with fluorescence, UV–vis and circular dichroism spectroscopies. The values of van’t Hoff enthalpy calculated from the temperature dependence of the binding constant do not agree with the calorimetric enthalpies indicating temperature dependent...

A novel class of thymine, adenine and lipoamino acid based non-viral carriers for gene delivery has been developed. Their ability to bind to DNA by hydrogen bonding was confirmed by NMR diffusion, isothermal titration calorimetry and transmission electron microscopy experiments.

A novel technique to estimate the distribution state of carboxyl groups within submicron-sized, carboxylated polymer particles was proposed. For the purpose, carboxyl groups in two kinds of butyl methacrylate-methacrylic acid copolymer emulsion, which were prepared by emulsion copolymerizations with different monomer addition methods, were titrated by isothermal titration calorimetry with an ex...

DNA double helices comprising two neighbouring artificial imidazole-Ag(I)-imidazole base pairs incorporate the Ag(I) ions in a cooperative fashion. Isothermal titration calorimetry data confirm the theoretically predicted binding stoichiometry and indicate that Ag(I) binds with association constants >10(6) M(-1) to the metal-free DNA duplex.

The host-guest interaction of a series of cyclohexyl-appended guests with cucurbit[8]uril (Q[8]) was studied by ¹H NMR spectroscopy, isothermal titration calorimetry (ITC), and X-ray crystallography. The X-ray structure revealed that two cycloalkane moieties can be simultaneously encapsulated in the hydrophobic cavity of the Q[8] host to form a ternary complex for the first time.
