BACKGROUND Vaccine development in human Plasmodium falciparum malaria has been hampered by the exceptionally high levels of CD8(+) T cells required for efficacy. Use of potently immunogenic human adenoviruses as vaccine vectors could overcome this problem, but these are limited by preexisting immunity to human adenoviruses. METHODS From 2007 to 2010, we undertook a phase I dose and route find...

BACKGROUND Recent studies have associated telomerase with prognostic factors and survival in neuroblastoma. PROCEDURE We examined telomerase activity by telomere repeat amplification protocol (TRAP) and expression of the RNA component of telomerase (hTR) by Northern blotting in 106 primary neuroblastoma tumors and 22 established cell lines. RESULTS Overall survival at 5 years for all 106 tu...

Coactivators previously implicated in ligand-dependent activation functions by thyroid hormone receptor (TR) include p300 and CREB-binding protein (CBP), the steroid receptor coactivator-1 (SRC-1)-related family of proteins, and the multicomponent TR-associated protein (TRAP) complex. Here we show that two positive cofactors (PC2 and PC4) derived from the upstream stimulatory activity (USA) cof...

Due to its 3 carbonic acid groups being available for bioconjugation, the TRAP chelator (1,4,7-triazacyclononane-1,4,7-tris(methylene(2-carboxyethylphosphinic acid))) is chosen for the synthesis of trimeric bioconjugates for radiolabelling. We optimized a protocol for bio-orthogonal TRAP conjugation via Cu(I)-catalyzed Huisgen-cycloaddition of terminal azides and alkynes (CuAAC), including a de...

In May 2014, the Rosetta spacecraft is scheduled to rendezvous with the comet Churyumov-Gerasimenko ('67P'). One of the instruments on board the 'Lander' which will descend on to the surface of the comet is a miniaturised GC/MS system that incorporates an ion trap mass spectrometer, specially developed for isotope ratio analysis. This article describes the development and optimisation of the io...

Energy-dependent protein degradation is studied through the dual bead ClpXP motility assay. Processing of folded proteins involves recognition, unfolding, translocation, and degradation stages. A dual optical trap, in a passive force-clamp geometry, exhibits bead-to-bead displacements that directly follow subprocesses underlying protein degradation. Discrete nanometer-scale displacements of the...

Different types of sediment trap have been introduced so far to be used in erosion measuring stations, each with its own advantages and disadvantages in trapping aeolian particles. These traps have different collection and retention efficiency due to aerodynamic conditions. Some characteristics such as isokinetic feature of the trap, collection efficiency, time needed to download sediment, capa...

Introduction & Objective : Telomerase is a reverse transcriptase enzyme that is required in immortalized cancer cells. Early detection of telomerase activity in tumors particularly in bladder cancer is promising. The aim of this study was to compare specificity and sensitivity between cytology and telomerase activity in diagnosis of bladder cancer. Materials & Methods : This study was a case...

A simple, quick and sensitive method was used to detect telomerase activity in Plasmodium falciparum. The telomeric repeat amplification protocol (TRAP assay) was modified using electrophoresis and staining with SYBR-green I to detect telomerase activity in a range of 10 to 10(7) parasites. This might be a useful way to ascertain telomerase activity in different types of nontumor cells.
