We have synthesized and characterized a water-soluble gold(III) corrole (1-Au) that is highly toxic to cisplatin-resistant cancer cells. Relative to its 1-Ga analogue, axial ligands bind only weakly to 1-Au, which likely accounts for its lower affinity for human serum albumin (HSA). We suggest that the cytotoxicity of 1-Au may be related to this lower HSA affinity.
The human serum albumin (HSA) secondary structure modifications induced by the chrysotile surface have been investigated via computational molecular dynamics (MD) and experimental infrared spectroscopy (FTIR) on synthetic chrysotile nanocrystals coated with different amount of HSA. MD simulations, conducted by placing various albumin subdomains close to the fixed chrysotile surface, show an ini...
Defining the interaction of newly synthesized compounds with plasma proteins is an important step in drug development process. Chromatographic techniques can be successfully used predicting biopharmaceutical and pharmacokinetic properties compounds. The aim this study to investigate isolate most molecular that affect 20 chalcones commercial (lopinavir, ritonavir, darunavir ivermectin) human ser...
The reaction between cisplatin and human serum albumin (HSA) was investigated by X-ray crystallography and crystal structures of the cisplatin/HSA adduct were eventually solved for the first time. Structural data unambiguously prove that cisplatin mainly binds to His105 and Met329 side chains; additional binding sites are detected at His288, Met298, and Met548 and at His535, His67 and His247.
In this paper, the quartz crystal microbalance with dissipation monitoring (QCM-D) was used to investigate hydrophobicity and binding strength (KD) for 10 different drugs interacting human serum albumin (HSA). Quantitative structure activity relationship (QSAR) analysis determine between drug (ClogP) HSA log(1/KD). The results are compared prior knowledge on bovine (BSA) binding. We demonstrate...
Studies were performed to elucidate the mechanism responsible for the reduction in Km values of UDP-glucuronosyltransferase 2B7 (UGT2B7) substrates observed for incubations conducted in the presence of albumin. Addition of bovine serum albumin (BSA) and fatty acid-free human serum albumin (HSA-FAF), but not "crude" HSA, resulted in an approximate 90% reduction in the Km values for the glucuroni...
Human serum albumin (HSA), bovine serum albumin (BSA) and human IgG were immobilized on latex beads to detect cell surface components of Helicobacter pylori binding to BSA, HSA, and IgG by a particle agglutination assay (PAA). In a total of 32 H. pylori strains tested, 16 strains interacted with BSA and 12 strains with HSA; 24 strains expressed binding of human IgG. The specificity of the agglu...
WANG, Y.F., CHUANG, M.H., CHIU, J.S., CHAM, T.M. and CHUNG, M.I. On-Site Preparation of Technetium-99m Labeled Human Serum Albumin for Clinical Application. Tohoku J. Exp. Med., 2007, 211 (4), 379-385 ── Technetium-99m labeled human serum albumin (Tc-99m HSA) is an important radiopharmaceutical for clinical applications, such as cardiac function tests or protein-losing gastroenteropathy assessm...
A monomorphic albumin-like protein (CfSA) has been purified to homogeneity from the serum of African air-breathing catfish Clarias gariepinus Bloch. It has a molecular mass of approximately =70 kD and shows a lesser electrophoretic mobility than human serum albumin (HSA) in native gels. The protein exhibits cross-reactivity against rabbit anti-HSA serum and shows considerable similarity with HS...
