A Real-time polymerase chain reaction (PCR) with melting analysis was devised to target bacterial and fungal genes together with the most prevalent antimicrobial resistance genes in 250 positive blood culture broths. This method allowed the blood culture cultivated pathogens to be classified into clinically relevant groups such as Enterobacteriaceae, oxidase-positive bacilli, oxidase-positive c...

Microbiological methods such as Gram staining and culture of low respiratory tract secretions are necessary to detect the real bacterial pathogens that cause lower respiratory tract infections (LRTI). In many cases, Gram stain analysis may be unreliable to determine the cause of LRTI, and bacterial cultures provide adequate information. However, we herein report two cases of lower respiratory t...

An 85-year-old woman was admitted to our hospital for steroid therapy for relapsing nephrotic syndrome. During hospitalization, she complained of sudden epigastric pain at night. Although there were signs of peritoneal irritation, CT showed a large amount of ascitic fluid, but no free intraperitoneal gas. Gram staining of ascitic fluid obtained by abdominal paracentesis showed Gram-negative rod...

Figure 1. Gram stain of the isolate from a blood culture (original magnification, 1000x) showing gram-negative rods with both filamentous (arrow) and dumbbell-shaped (arrowheads) morphologies characteristic of Cardiobacterium hominis. Diagnosis: Cardiobacterium hominis prosthetic valve endo-carditis. C. hominis is one of the fastidious gram-negative bacteria that belong to the HACEK group of or...

We reviewed 194 revision arthroplasties of the hip and knee performed over a ten-year period. The results of intraoperative Gram staining were available in 169 (87%). Thirty-two were found to be infected (11 hips and 21 knees) and 137 had no evidence of infection. Intraoperative Gram staining was negative in all 169 cases. The method therefore had a sensitivity of 0% for detecting infection. We...

This paper deals with an investigation of the effect of these reactions on bacteria. Not only the common staining behavior but also certain other similarities (Fischer et al., 1949; Fischer, 1951) between intact wool and gram negative bacteria, medium degraded wool and gram variable bacteria, as well as between highly degraded wool and gram positive bacteria suggested that reversal of gram stai...

Application of silicate solutions to living or heat-killed pneumococci and to certain "viridans" streptococci causes their conversion from a Gram-positive to a Gram-negative state. The original staining properties can be restored by suspending the silicate-treated bacteria in alkaline solutions of various salts but not by simple washing in water. Living pneumococci and the strains of streptococ...

OBJECTIVE To evaluate Helicobacter pylori (H. pylori) detection by histological staining methods, and to compare with those of Gram staining and polymerase chain reaction (PCR). METHODS This is a cross-sectional study conducted at the Department of Microbiology, Shree P. M. Patel Paramedical College, Anand, Gujarat, India on 436 patients attending the Deep Surgical Hospital, Anand, Gujarat be...

A total of 803 strains of gram-negative nonfermentative and oxidase-positive fermentative bacteria (38 taxa) were investigated for their ability to hydrolyze 53 different fluorogenic 4-methylumbelliferyland 1-naphthylamide-linked substrates within 6 h of incubation. The hydrolysis of 16 fluorogenic substrates showed high separation index values among the tested taxa, was reproducible, and showe...
