1. In a search for suitable tissues for the cultivation of yellow fever virus in vitro, mouse embryos were inoculated with this virus in utero. A titration for virus content of the various organs of the embryos indicated that the virus was present in the brain in greatest concentration. 2. Unmodified strains of yellow fever virus were readily adapted to cultivation in vitro in a medium consisti...

background and aims: infectious bursal disease (ibd) is an acute contagious viral disease of birds worldwide. the causative virus induces a persistent immune suppression following destroy b lymphocytes precursors in bursal lymphoid follicles. vaccination is the main strategy for prevention of the disease in commercial poultry industry. materials and methods: to produce a live vaccine against th...

A microneutralization test in PK(15) cells was developed to measure the neutralizing antibody response of a group of ponies experimentally challenged with louping ill virus. Viral cytopathic effect was maximal after 6 days of incubation, at which point titration endpoints were clear-cut and readily determinable. The assay compared favorably with the mouse neutralization test for accuracy and ea...

Two critical steps when investigating an animal model of a virus infection are consistently successfully infecting animals and accurately determining viral titers in tissue throughout the course of infection. Here we discuss in detail how to infect mice with SARS-CoV and then quantify the titer of virus in the lung.

Sheeppox virus from an outbreak of sheeppox that occurred in Srinagar (Jammu and Kashmir, India) in 2000 was isolated by inoculation of susceptible sheep and further re-isolated in cell culture. The field virus, adapted to grow in lamb testes culture, was evaluated for its potential use as challenge virus in potency testing of sheeppox vaccine currently in use. The virus (passage 6) produced se...

The complement fixation elution profiles of dengue-2 virus-infected suckling mouse brain supernatant fluids from Sephadex G-200 columns were compared by the antigen end-point titration procedure and by a complement dilution technique. The latter technique was found to be a quick method for approximating the complement-fixing antigen in eluates.

background and aims: infectious laryngotracheitis virus (iltv) infection causes a highly contagious respiratory disease in poultry leads to significant economic losses in the poultry industry. the genomic and antigenic characteristics between virulent and vaccine strains of iltv are very similar. the ability of embryonated chicken neural stem (cns) cell for differential diagnosis of iltv strain...

A semiautomated method of an indirect immunofluorescence technique for the titration of antibodies directed against viral capsid antigens of Epstein-Barr virus has been developed with Microtiter system units. By this method, a technician is able to titrate some hundred samples daily. The technique is safe, easy, and reproducible. The various procedures are described, and the sensitivity of the ...

An automatic method, utilizing the AutoAnalyzer, for titration of influenza virus haemagglutination inhibitors is described. The titres obtained compare favourably with those obtained by densitometric methods. So far, the technique has not proved superior to the densitometric methods in either speed of operation or in reproducibility but various developments are suggested to effect this.

To simplify the titration of infectious varicella-zoster virus (VZV), we generated a reporter cell line that produced luciferase in a dose-dependent manner upon infection with cell-free VZV. A few VZV-infected cells were detectable by coculturing with the cell line. We demonstrated the usefulness of the cell line for antiviral studies.
