Arylboronic acids were found to be strong competitive inhibitors of subtilisin and chymotrypsin. The binding constants are strongly pH dependent and give a Hammett-type plot with a slope of -0.885. The pH dependence, the Hammett plot, and nmr model-system studies indicate that inhibition is due to electron-pair donation by the active site histidine to the bound inhibitor.

Second-order rate constants (kN) have been measured spectrophotometrically for the nucleophilic displacement reactions of 4-nitrophenyl X-substituted-cinnamates (4a-4e) and Y-substituted-phenyl cinnamates (5a-5e) with Z-substituted-phenoxide anions in 80 mol % H2O/20 mol % DMSO at 25.0 ± 0.1 C. The Hammett plot for the reactions of 4a-4e with 4-chlorophenoxide (4-ClPhO) consists of two intersec...

Negative-ion electrospray ionisation mass spectrometry with an anionic phosphine ligand enables detection of key intermediates in the Sonogashira reaction. MS/MS techniques are used to generate a Hammett plot for the key reductive elimination step.

A "V"-shaped Hammett plot shows that resonance-assisted hydrogen bonding does not dictate the strength of the intramolecular hydrogen bond in the E isomers of hydrazone-based switches because it involves an aromatic pyridyl ring.

A kinetic study on aminolysis of 4-chloro-2-nitrophenyl X-substituted-benzoates (6a-i) in MeCN is reported. The Hammett plot for the reactions of 6a-i with piperidine consists of two intersecting straight lines, while the Yukawa-Tsuno plot exhibits an excellent linear correlation with ρX = 1.03 and r = 0.78. The nonlinear Hammett plot is not due to a change in rate-determining step (RDS) but is...

Measurements are reported on the kinetics of the lysozyme-catalysed hydrolysis of several beta-aryl di-N-acetylchitobiosides, some of which have been synthesized for the first time. The catalytic rate constants (k(cat.)) at 45 degrees yield a curved Hammett plot (concave up) and the plot of DeltaH(double dagger) versus DeltaS(double dagger) has a sharp break. Substrates with electron-withdrawin...

Hydrogen bond strength in host-guest systems is modulated by many factors including preorganization, steric effects, and electronic effects. To investigate how electronic effects impact barbiturate binding in bifurcated Hamilton receptors, a library of receptors with differing electronic substituents was synthesized and (1)H NMR titrations were performed with diethyl barbital. The Hammett plot ...

[structure: see text] The trans/cis ratio of the amide bond in N-formylproline phenylesters correlates with electron withdrawal by a para substituent. The slope of the Hammett plot (rho = 0.26) is indicative of a substantial effect. This effect arises from a favorable n --> pi interaction between the amide oxygen and ester carbonyl. In a polypeptide chain, an analogous interaction can stabilize...

Evaluation of a new proton sponge, bis-1,1,8,8-(tetramethylguanidino)naphtalene (TMGN), in substituted phenols O-alkylation by methyl iodide in DMF has been studied. Kinetic measurements were performed in N,N-dimethylformamide-d7 and followed by H NMR using stoichiometric amounts of reagents. Plot of the results shows that the reaction follows an almost perfect second order rate law. However th...

A series of C4 substituted alpha-ketooxazoles were examined as inhibitors of the serine hydrolase fatty acid amide hydrolase in efforts that further define and generalize a fundamental substituent effect on enzyme inhibitory potency. Thus, a plot of the Hammett sigma(m) versus -logK(i) provided a linear correlation (R(2)=0.90) with a slope of 3.37 (rho=3.37), that is of a magnitude that indicat...
