white-rot basidiomycetes are one of the most important lignolytic microorganisms. these fungi have been reported to secrete three main classes of lignin degrading enzymes: lignin peroxidases (lips), manganese peroxidases (mnps) and laccases. in this study, for the first time the lignin degrading capability of two plant pathogens i.e. hyphodermella rosae and h. corrugata was evaluated using both...

Chance (1) has proposed that the peroxidase-catalyzed oxidation of diiydroxyfumaric acid proceeds in two steps. The first step is the oxidation of the hydrogen donor by oxygen, the reaction being catalyzed by a manganese-activated peroxidase and accompanied by the production of hydrogen peroxide. The second step is the oxidation of additional hydrogen donor by hydrogen peroxide, this reaction a...

The requirement for manganese in peroxidase-catalyzed oxidations has hitherto been considered to be very specific. Various investigators (l-6) have tested a number of other ions (Zn, Mg, Cu, Co, Fe, Ni, Cd, Ca, Al, Sn, and molybdate) in different peroxidase-catalyzed oxidations, but with the exception of a slight stimulation of reduced diphosphopyridine nucleotide oxidation by cobaltous ions, t...

Phanerochaete chrysosporium is a white rot fungus which secretes a family of lignin-degrading enzymes under nutrient limitation. PSBL-1 is a mutant of this organism that generates the ligninolytic system under nonlimiting conditions during primary metabolism. Lignin peroxidase, manganese peroxidase, and glyoxal oxidase activities for PSBL-1 under nonlimiting conditions were 4- to 10-fold higher...

Apoproteins of cytochrome c peroxidase, horseradish peroxidase, and sperm whale myoglobin were recombined with manganese complexes of proto-, hemato-, meso-, and deuteroporphyrins to form manganese porphyrin-protein complexes. These complexes were purified by column chromatography. All the manganese porphyrin-containing cytochrome c peroxidases were crystallized. Light absorption maxima of mang...

The white rot fungus Phanerochaete chrysosporium has significant pollutant-degrading capabilities with its oxidoreductases. The lignin-degrading enzyme system of this fungus also allows the breakdown of different organic pollutants (xenobiotics). The most significant enzymes in these processes are the lignin peroxidase and the manganese peroxidase. In this study, Phanerochaete chrysosporium str...

In this study, we report the manganese peroxidase production ability from a Fusarium sp. strain using an inexpensive medium of agriculture residues of either rice straw or wood chips as carbon source. The highest manganese peroxidase activity on rice straw medium and on wood chips was 1.76 U/mL by day 9 and 1.91 U/mL by day 12, respectively.

Under secondary metabolic conditions the white rot basidiomycete Phanerochaete chrysosporium mineralizes 2,4-dichlorophenol (I). The pathway for the degradation of 2,4-dichlorophenol (I) was elucidated by the characterization of fungal metabolites and of oxidation products generated by purified lignin peroxidase and manganese peroxidase. The multistep pathway involves the oxidative dechlorinati...

A cDNA clone encoding a manganese-dependent peroxidase from the filamentous fungus Phanerochaete chrysosporium was isolated and characterized. The clone, AMP-1, was isolated by screening a X g t l l expression library with polyclonal antibodies raised against a purified manganese-dependent peroxidase (isozyme H4, PI 4.5). The XMP-1 cDNA sequence predicts a mature protein containing 358 amino a...

The upstream sequences of all published lignin peroxidase and manganese peroxidase genomic clones from Phanerochaete chrysosporium were analyzed. This analysis revealed the presence of putative activator protein-2 (AP-2) recognition sequences in 11 of 15 lignin peroxidase genes. The lignin peroxidase clone GLG6 and the manganese peroxidase gene (mnp-1) have two copies of putative AP-2 sequence ...
