Canine DNA samples from South Africa were found to contain 16S rRNA gene nucleotide and citrate synthase gene nucleotide and deduced amino acid sequences that were most similar to Anaplasma phagocytophilum: 98%, 66%, and 69% similarity, respectively. This suggests that a new Anaplasma species closely related to A. phagocytophilum occurs in Africa.

The Tm value of DNA from Anaplasma centrale and Anaplasma marginale was found to be 87.1 degrees C and 89.3 degrees C, respectively. The G + C content, calculated from the Tm, was 45.1% for A. centrale and 48.5% for A. marginale. Identical hybridization patterns were obtained when the DNA from one species was hybridized to restriction endonuclease-digested DNA from the other species.

We developed PCR-based assays to distinguish a human pathogenic strain of Anaplasma phagocytophilum, Ap-ha, from Ap-variant 1, a strain not associated with human infection. The assays were validated on A. phagocytophilum-infected black-legged ticks (Ixodes scapularis) collected in Canada. The relative prevalence of these 2 strains in I. scapularis ticks differed among geographic regions.
