Formylglycine-generating enzyme (FGE) is an O2 -utilizing oxidase that converts specific cysteine residues of client proteins to formylglycine. We show that CuI is an integral cofactor of this enzyme and binds with high affinity (KD =of 10-17  m) to a pair of active-site cysteines. These findings establish FGE as a novel type of copper enzyme.

Fluorophore labeling of proteins while preserving native functions is essential for bulk Förster resonance energy transfer (FRET) interaction and single molecule imaging analysis. Here we describe a versatile, efficient, specific, irreversible, gentle and low-cost method for labeling proteins with fluorophores that appears substantially more robust than a similar but chemically distinct procedu...

The loss functions of deep neural networks are complex and their geometric properties are not well understood. We show that the optima of these complex loss functions are in fact connected by a simple polygonal chain with only one bend, over which training and test accuracy are nearly constant. We introduce a training procedure to discover these high-accuracy pathways between modes. Inspired by...

conclusions the results suggest that fge can improve the antibiotic sensitivity of these pathogens to some antibiotics. results fresh garlic extract displayed evident inhibition properties against c. albicans and mrsa, yet weak inhibition properties against p. aeruginosa. additionally, fge showed the potential to improve the effect of antibiotics on antibiotic resistant pathogens. the synergism...

BACKGROUND Recently, magnetic resonance (MR)-conditional implantable cardioverter defibrillator (ICD) systems have become available. However, associated cardiac MR image (MRI) quality is unknown. The goal was to evaluate the image quality performance of various cardiac MR sequences in a multicenter trial of patients implanted with an MR-conditional ICD system. METHODS AND RESULTS The Evera-MR...

Formylglycine generating enzyme (FGE) performs a critical posttranslational modification of type I sulfatases, converting cysteine within the motif CxPxR to the aldehyde-bearing residue formylglycine (FGly). This concise motif can be installed within heterologous proteins as a genetically encoded "aldehyde tag" for site-specific labeling with aminooxy- or hydrazide-functionalized probes. In thi...

The properties of therapeutic proteins can be enhanced by chemical modification. Methods for site-specific protein conjugation are critical to such efforts. Here, we demonstrate that recombinant proteins expressed in mammalian cells can be site-specifically modified by using a genetically encoded aldehyde tag. We introduced the peptide sequence recognized by the endoplasmic reticulum (ER)-resid...

Recently, numerous studies have demonstrated the effectiveness of a segmented inversion recovery fast gradient echo (seg IR-FGE) sequence for differentiating injured from normal myocardium. This technique for delayed enhancement imaging has been shown to be effective in identifying the presence and extent of myocardial infarction, as well as predicting improvement in contractile function after ...

With the development of heterogeneous network structure, dynamic user requests as well complex service types and applications scenarios, current networks may not accommodate increasingly stringent requirements. As a result, research beyond fifth generation (B5G) or sixth (6G) has been put on agenda. In B5G/6G networks, achieving automatic, flexible, cost-effective orchestration management resou...

OBJECTIVE To test the hypothesis that eustachian tube opening efficiency, measured as the fractional gradient equilibrated (FGE), is lower in 6-year-old children with no middle ear disease but a well-documented history of recurrent acute otitis media, as compared with children with a negative disease history (control). STUDY DESIGN Cross-sectional study. SETTING Tertiary care pediatric hosp...
