Tacrolimus (formerly known as FK506) is a macrolide antibiotic isolated from the soil fungus Strtptomyus tsukubamsis. 1 Although it is totally distinct in molecular structure from cyclosporine A (CsA), a cyclic endecapeptide extracted from the fungus Tolypociadium inflatum, the two immunosuppressants share a remarkably similar, selective inhibitory action on the activation and proliferation of ...
The macrocyclic polyketide tacrolimus (FK506) is a potent immunosuppressant that prevents T-cell proliferation produced solely by Streptomyces species. We report here the first draft genome sequence of a true FK506 producer, Streptomyces tsukubaensis NRRL 18488, the first tacrolimus-producing strain that was isolated and that contains the full tacrolimus biosynthesis gene cluster.
We describe the first reported case of a syndrome of inappropriate secretion of antidiuretic hormone (SIADH) induced by low-dose tacrolimus in a patient with autoimmune disease. A 41-year-old man with systemic lupus erythematosus (SLE) developed hyponatremia induced by SIADH after administration of tacrolimus (0.06 mg/kg per day). In this case, the hyponatremia promptly resolved upon withdrawal...
Syndrome of inappropriate antidiuretic hormone secretion (SIADH) is an important electrolyte abnormality that can occur following allogeneic stem cell transplantation. We herein report the case of a 20-year-old man who developed SIADH three weeks after undergoing cord blood transplantation. Tacrolimus administration was suspected to be a cause of the disorder. In addition to restricting water i...
tacrolimus is a main drug for induction and maintenance immunosuppression for patients with kidney transplants in many centers. one of important side effect of drug is post-transplant diabetes mellitus .of course; diabetes ketoacidosis (dka) is rare. in this report, a 12-year-old girl with dka was presented after 45 days of kidney transplantation.
Topical 0.03% tacrolimus was used for treatment of a Korea Jindo dog diagnosed with pemphigus erythematosus. The dog was slowly improved following application of tacrolimus but did not achieve complete remission until end of this study. No adverse effects on clinical or laboratory parameters were noted during the topical tacrolimus therapy period.
Tacrolimus (FK506) and cyclosporine (cyclosporin A, CsA) are cornerstone immunosuppressive agents administered to solid organ transplant recipients to prevent and treat allograft rejection. The discovery of cyclosporine in the 1970s, and its entry into the collection of immunosuppressants in the early 1980s, was a major breakthrough in medicine. Cyclosporine was the most successful antirejectio...
The article deals with the contribution of tacrolimus (Prograf) to improvement in kidney transplant results. Tacrolimus, in comparison with cyclosporine significantly reduces the incidence of acute rejection and improves survival of grafts as well as patients. Based on the literature, the primary immunological differences between tacrolimus and cyclosporine effects are pointed out. These differ...
Introduction Renal transplantation is the therapeutic of choice for the end stage renal disease. With advanced immunosuppressive drug, the outcome of renal transplantation becomes favorable in the present day (1). Tacrolimus is a widely used immunosuppressive drug for post-renal transplantation (2). The concentration of tacrolimus is important for success in management of the patient. The effec...
immunosuppressants require therapeutic drug monitoring because of their narrow therapeutic index and significant inter-individual variability in blood concentrations. this variability can be because of factors like drug-nutrient interactions, drug-disease interactions, renal-insufficiency, inflammation and infection, gender, age, polymorphism and liver mass. drug monitoring is widely practiced ...
