Abstract Background and Aims IgA nephropathy (IgAN) is the most common primary glomerular disease in children adolescents worldwide, one of major causes end-stage renal China. Minimal change disease(MCD) pathological type nephrotic syndrome children. In some rare cases, pathology these patient has deposits mesangial regionns diffuse fusion podocyte foot process,which called IgA-MCD. Previous studies have analyzed characteristics IgA-MCD from aspects clinical manifestations, features, therapeutic response prognosis, but molecular mechanism its occurrence development not been clarified. For first time, single-cell transcriptome sequencing technology was used to compare IgA-MCD, IgAN MCD, order describe unique elucidate related manifestations. It lays foundation for formulating more appropriate treatment plan improving prognosis. Method Renal fine-needle puncture tissues were collected 3 with IgAN, 4 1 child Children's Hospital Chongqing Medical University, adjacent normal tumor as control group. Children three groups had manifestations syndrome, nephrotic-level proteinuria at time sampling. High-throughput, unbiased performed on above tissue samples, results systematically compared analyzed. Results By analyzing data, we described cell maps form albuminuria different pathologic types (IgAN, IgA-MCD). Twelve types, such mesenchymal cells podocytes, defined by classical marker genes. comparing constituent ratio PC analysis subsets group groups, it found that closer MCD. differential genes those group, number IgAN’s podocytes significantly higher than other two groups. Compared HMGCS2 up-regulated cellular energy metabolism enhanced. Through pairwise comparison among there no significant difference between CXCL12 gene expression up-regulated, can recruit immune lead damage. Conclusion The overall transcription profile similar specifically highly expressed which may be a molecule functional changes podocyte. up-regulation CD81 signal leads activation, proliferation, secretion extracellular matrix IgAN-MCD cells.We associations differences MCD level, provided new perspective insight follow-up study pathogenesis diseases.
