Acute Promyelocytic Leukemia (APL): A Review of the Classic and Emerging Target Therapies towards Molecular Heterogeneity

چکیده
The occurrence of severe bleeding syndrome because the PML-RARα fusion protein is a life-threatening event in APL. This destabilizes homeostasis, maturation, remodeling, and tissue regeneration addition to hampering maintenance differentiation hematopoietic cells into different lineages, fixing promyelocyte stage. APL classic example how effective targeted therapy and, therefore, important use such overall survival patients, which this case represented by ATRA/ATO. Despite that, about 10% cases patients demonstrate resistance treatment. Facing scenario, we point out promising target therapies as those recommended NCCN Leukemia Net. Since heterogeneous molecular disease, it great importance understand combined chemotherapy, therapy, immune-based are these patients.

Acute promyelocytic leukemia: a model of molecular target based therapy.
Leukemia, a group of hematological malignancies characterized by clonal expansion of hematopoietic cells with uncontrolled proliferation, decreased apoptosis and blocked differentiation, is one of the most notorious enemies of mankind which accounts for some 300,000 new cases and 222,000 deaths each year worldwide. Leukemia can be divided into acute or chronic, lymphoid or myeloid types, based ...

The outcome of adults and children with Acute Promyelocytic Leukemia (APL) has dramatically changed since the introduction of all trans retinoic acid (ATRA) therapy. Based on the results of several multicenter trials, the current recommendations for the treatment of patients with APL include ATRA and anthracycline-based chemotherapy for the remission induction and consolidation, and ATRA combin...

Since the first description of acute promyelocytic leukemia (APL) in 1957 as the most malignant form of acute leukemia, several developments have paved the way to make this disease the most curable leukemia in adults and change the paradigm of cancer treatment. Therapy of APL was pioneered by Bernard et al in 1973 demonstrating a striking sensitivity to daunorubicin, probably related to signifi...
