Cell-Main Spectra Profile Screening Technique in Simulation of Circulating Tumour Cells Using MALDI-TOF Mass Spectrometry

Circulating atypical cells (CAC) are released from a primary tumour site into peripheral blood and indicators of cancer metastasis. CAC occur at very low frequency in circulating blood, their detection remains challenging. Moreover, white (WBC) the major contaminant enriched samples. Here, we developed matrix-assisted laser desorption ionization–time flight mass spectrometry (MALDI-TOF MS) as novel characterization platform. Main spectra profiles (MSP) normal were generated by MALDI-TOF MS, cell-main database was then compiled analysed using MALDI Biotyper software. Logarithmic scores accurately predicted distinct cell types. The feasibility this workflow validated simulated samples, which prepared 5000 WBC three healthy individuals spiked with varying numbers (3, 6, 12, 25, 50, 100) lung, colon, or prostate cells. MS able to detect down six over background noise significantly higher predictive compared alone. Further development cell-MSP cover all types sourced lines patient tumours may enable use cancer-screening platform clinical settings future.
