Central carbon metabolite profiling reveals vector-associated differences in the recombinant protein production host Escherichia coli BL21

The Gram-negative bacterium Escherichia coli is the most widely used host for recombinant protein production, both as an industrial expression platform and a model system at laboratory scale. production industry generates proteins with direct applications biopharmaceuticals in technological processes central to plethora of fields. Despite increasing economic significance importance E. organism, only few studies have focused on carbon metabolic landscape during high-level production. In present work, we applied four targeted CapIC- LC-MS/MS methods, covering over 60 metabolites, perform in-depth metabolite profiling effects strains derived from BL21, carrying XylS/ Pm vectors different characteristics. mass-spectrometric was complemented study growth kinetics batch bioreactors. Our work shows robustness metabolism when introducing increased plasmid copy number, well greater induction challenge, especially strong promoters are used.
