چکیده

Bombyx mori nucleopolyhedrovirus (BmNPV) is a kind of pathogen that causes huge economic losses to silkworm production. Although Autographa californica (AcMNPV) and BmNPV are both baculoviruses, the host domains these two viruses have almost no intersection in nature. Recently, it has been found some silkworms could be infected by recombinant AcMNPV through puncture, which provided valuable material for studying infection mechanism baculovirus silkworm. In this study, comparative transcriptomics was used analyse hemolymph differentially resistant strains following inoculation. There were 678 DEGs p50 515 C108 viral infection. Among them, upregulation downregulation similar p50; however, upregulated nearly twice as numerous downregulated C108. The different differed GO enrichment. Based on KEGG enrichment, mainly enriched metabolic pathways apoptosis pathway Moreover, 13 genes involved 11 analysed. apoptosis, function BmTex261 showed highest expression significant response C108, indicating anti-AcMNPV This further validated significantly decreased gene lef3 after overexpression BmN cells. results provide theoretical reference molecular resistance silkworms.

Gene transduction in mammalian cells using Bombyx mori nucleopolyhedrovirus assisted by glycoprotein 64 of Autographa californica multiple nucleopolyhedrovirus

Autographa californica multiple nucleopolyhedrovirus (AcMNPV), an alphabaculovirus, has been widely utilized for protein expression in not only insect cells but also mammalian cells. AcMNPV is closely related to Bombyx mori nucleopolyhedrovirus (BmNPV), and nucleotide sequences of AcMNPV genes have high similarity with those of BmNPV. However, the transduction of BmNPV into mammalian cells has ...

Comparative Subcellular Proteomics Analysis of Susceptible and Near-isogenic Resistant Bombyx mori (Lepidoptera) Larval Midgut Response to BmNPV infection

The molecular mechanism of silkworm resistance to Bombyx mori nucleopolyhedrovirus (BmNPV) infection remains largely unclear. Accumulating evidence suggests that subcellular fractionation combined with proteomics is an ideal technique to analyse host antiviral mechanisms. To clarify the anti-BmNPV mechanism of the silkworm, the near-isogenic line BC9 (resistant strain) and the recurrent parent ...

The Differential Expression of BmGlcNAcase2 in Strains of Bombyx mori (Lepidoptera: Bombycidae) With Different Susceptibility to Bombyx mori (Lepidoptera: Bombycidae) Nucleopolyhedrovirus Infection

GlcNAcase is a glycosyl hydrolase located in the lysosomes of numerous organisms. Levels of the protein, β-N-acetylglucosaminidase 2 (GlcNAcase2), which is a member of the GlcNAcase family, are different in two strains of the silkworm Bombyx mori that have different resistance to Bombyx mori nucleopolyhedroviruses (BmNPVs). We identified six single-nucleotide differences in the GlcNAcase2 codin...

Lymantria dispar nucleopolyhedrovirus hrf-1 expands the larval host range of Autographa californica nucleopolyhedrovirus.

The gypsy moth (Lymantria dispar) is nonpermissive for Autographa californica nucleopolyhedrovirus (AcNPV) infection. We previously isolated a gene, host range factor 1 (hrf-1), from L. dispar nucleopolyhedrovirus that promotes AcNPV replication in Ld652Y cells, a nonpermissive L. dispar cell line (S. M. Thiem, X. Du, M. E. Quentin, and M. M. Berner, J. Virol. 70:2221-2229, 1996). In the presen...

The ecdysteroid UDP-glucosyltransferase gene of Autographa californica nucleopolyhedrovirus alters the moulting and metamorphosis of a non-target insect, the silkworm, Bombyx mori (Lepidoptera, Bombycidae).

The Autographa californica nucleopolyhedrovirus (AcMNPV) does not infect the silkworm and molecular studies on silkworm insusceptibility have not been performed. In cultured cells of the silkworm, the expression of viral genes has been reported. The expression of AcMNPV genes and their effect in vivo and in vitro was studied. In this study, the early gene, the ecdysteroid UDP-glucosyltransferas...
