CPSF30-L-mediated recognition of mRNA m6A modification controls alternative polyadenylation of nitrate signaling-related gene transcripts in Arabidopsis

N6-methyladenosine (m6A), a ubiquitous internal modification of eukaryotic mRNAs, plays vital role in almost every aspect mRNA metabolism. However, there is little evidence documenting the m6A regulating alternative polyadenylation (APA) plants. APA controlled by large protein-RNA complex with many components, including CLEAVAGE AND POLYADENYLATION SPECIFICITY FACTOR30 (CPSF30). In Arabidopsis, CPSF30 has two isoforms and longer isoform (CPSF30-L) contains YT512-B Homology (YTH) domain, which unique to this study, we showed that CPSF30-L YTH domain binds vitro. cpsf30-2 mutant, transcripts genes several important nitrate signaling-related had shifts sites were correlated peaks, indicating these gene carrying tend be regulated APA. Wild-type could rescue defects metabolism cpsf30-2, but m6A-binding-defective mutants not. Taken together, our results demonstrated regulates Arabidopsis revealed reader affects signaling controlling APA, shedding new light on roles during RNA 3?-end processing
