Cre-Controlled CRISPR mutagenesis provides fast and easy conditional gene inactivation in zebrafish

چکیده
Abstract Conditional gene inactivation is a powerful tool to determine function when constitutive mutations result in detrimental effects. The most commonly used technique achieve conditional employs the Cre/loxP system and its ability delete DNA sequences flanked by two loxP sites. However, targeting with sites time labor consuming. Here, we show Cre-Controlled CRISPR (3C) mutagenesis circumvent these issues. 3C relies on gRNA Cre-dependent Cas9-GFP expression from same transgene. Exogenous or transgenic supply of Cre results subsequent interest. recombined cells become fluorescently visible enabling their isolation subjection various omics techniques. Hence, provides valuable alternative production loxP-flanked alleles. It might even enable multiple genes simultaneously should be applicable other model organisms amenable single integration transgenesis.

Supporting conditional mouse mutagenesis with a comprehensive cre characterization resource
Full realization of the value of the loxP-flanked alleles generated by the International Knockout Mouse Consortium will require a large set of well-characterized cre-driver lines. However, many cre driver lines display excision activity beyond the intended tissue or cell type, and these data are frequently unavailable to the potential user. Here we describe a high-throughput pipeline to extend ...

Existing transgenic RNA interference (RNAi) methods greatly facilitate functional genome studies via controlled silencing of targeted mRNA in Drosophila. Although the RNAi approach is extremely powerful, concerns still linger about its low efficiency. Here, we developed a CRISPR/Cas9-mediated conditional mutagenesis system by combining tissue-specific expression of Cas9 driven by the Gal4/upstr...

The International Knockout Mouse Consortium (IKMC) has produced a genome-wide collection of 15,000 isogenic targeting vectors for conditional mutagenesis in C57BL/6N mice. Although most of the vectors have been used successfully in murine embryonic stem (ES) cells, there remain a set of nearly two thousand genes that have failed to target even after several attempts. Recent attention has turned...
متن کاملGermline recombination by conditional gene targeting with Parvalbumin-Cre lines
Conditional gene targeting allows us to study gene function in specific tissues or cell types. This is commonly achieved by Cre DNA recombinase and its 34–base pair target sequences called loxP sites. Through the efforts of individual labs and largescale projects, a sizable collection of Cre mouse lines has been generated to express or delete specific genes in a wide range of cell types through...
متن کاملConditional gene knock-down by CRE-dependent short interfering RNAs.
Short interfering RNAs (siRNAs) are short (21-23 nt) double-stranded RNAs that direct the sequence-specific degradation of corresponding mRNAs, resulting in suppression of gene activity. siRNAs are powerful tools for gene functional analysis in mammals. Chemically synthesized siRNAs permit transient gene repression but preclude inhibition of stable gene products as well as long-term phenotypic ...
