Culture Methods to Study Apical-Specific Interactions using Intestinal Organoid Models

چکیده
The lining of the gut epithelium is made up a simple layer specialized epithelial cells that expose their apical side to lumen and respond external cues. Recent optimization in vitro culture conditions allows for re-creation intestinal stem cell niche development advanced 3-dimensional (3D) systems recapitulate composition organization epithelium. Intestinal organoids embedded an extracellular matrix (ECM) can be maintained long-term self-organize generate well-defined, polarized encompasses internal exposed basal side. This restrictive nature presents challenges accessing surface limits investigation biological mechanisms such as nutrient uptake host-microbiota/host-pathogen interactions. Here, we describe two methods facilitate access organoid support differentiation specific types. First, show how ECM removal induces inversion polarity generation apical-out 3D organoids. Second, 2-dimensional (2D) monolayers from single suspensions derived organoids, comprised mature differentiated These techniques provide novel tools study apical-specific interactions with cues promote use platform precision medicine.

Salmonella‐infected crypt‐derived intestinal organoid culture system for host–bacterial interactions
The in vitro analysis of bacterial-epithelial interactions in the intestine has been hampered by a lack of suitable intestinal epithelium culture systems. Here, we report a new experimental model using an organoid culture system to study pathophysiology of bacterial-epithelial interactions post Salmonella infection. Using crypt-derived mouse intestinal organoids, we were able to visualize the i...
متن کاملChallenges of Culturing Human Norovirus in Three-Dimensional Organoid Intestinal Cell Culture Models
Human noroviruses are the most common cause of acute gastroenteritis worldwide. Recently, cell culture systems have been described using either human embryonic intestinal epithelial cells (Int-407) or human epithelial colorectal adenocarcinoma cells (Caco-2) growing on collagen-I porous micro carrier beads in a rotating bioreactor under conditions of physiological fluid shear. Here, we describe...
متن کاملAdvancing Intestinal Organoid Technology Toward Regenerative Medicine
With the emergence of technologies to culture intestinal epithelial cells in vitro as various forms of intestinal organoids, there is growing interest in using such cultured intestinal cells as a source for tissue engineering and regenerative medicine. One such approach would be to combine the organoid technology with methodologies to engineer the culture environment, particularly the three-dim...
متن کاملRecognition for Culture Specific Interactions
The user’s behavior and his interpretation of interactions with others is influenced by his cultural background, which provides a number of heuristics or patterns of behavior and interpretation. This cultural influence on interaction has largely been neglected in HCI research due to two challenges: (i) grasping culture as a computational term and (ii) infering the user’s cultural background by ...
متن کاملFbxw7-associated drug resistance is reversed by induction of terminal differentiation in murine intestinal organoid culture
Colorectal cancer (CRC) is one of the top three cancer-related causes of death worldwide. FBXW7 is a known tumor-suppressor gene, commonly mutated in CRC and in a variety of other epithelial tumors. Low expression of FBXW7 is also associated with poor prognosis. Loss of FBXW7 sensitizes cancer cells to certain drugs, while making them more resistant to other types of chemotherapies. However, is...
