DEVELOPMENT OF METHOD FOR QUANTITATIVE DETERMINATION OF CEFQUINOME SULFATE IN PIGLETS BLOOD PLASMA USING HPLC/DMD

This manuscript presents a developed method for determining cefquinome sulfate in piglets blood plasma intended clinical and pharmaceutical research of veterinary drugs based on it. Blood proteins were precipitated twice with solution trichloroacetic acid. The supernatant was further purified by series solid-phase extraction. Separation performed an inverted phase Kinetex EVO С18 column using acetonitrile 0,1 % trifluoroacetic acide solusion as the mobile phase. gradient mode eluents used during 10 min at flow rate 1,4 ml/min. peak retention time is 4,2 min. specificity analytical checked comparing chromatographic separation sample enriched standard placebo. preparing loaded samples procedure building calibration graph described article. validation parameters “recovery” “coefficient variation” considered accordance criteria Council Directive 2002/657/EC. preparation fortified to construct manuscript. mean recovery from range 0.1-2.0 μg/ml cefquinom 102.3 %. linear concentration 0.1 – 4.0 sulfate. correlation coefficient determination 0.9998. results obtained study linearity this technique estimate correctness convergence. accuracy measurements evaluated examining known amounts analyte added control plasma.
