Development of New Oncolytic Virotherapy Targeting Breast Cancer Using Coxsackievirus B3

چکیده
Background/Aim: Breast cancer is the most common in women worldwide, and triple-negative breast (TNBC) highly refractory to current standard therapies. Oncolytic virotherapy has recently gathered attention as a new treatment candidate for cancers. Materials Methods: We previously developed Coxsackievirus B3 (CVB3) targeting lung cancers, demonstrated that miRNA target sequence insertion into CVB3 reduced its pathogenicity, retaining original oncolytic activity. In this study, we examined effects of against cells including TNBC cells. Results: infection killed time- titer-dependent manner, induced apoptosis. Nude mice transplanted with human were successfully treated both CVB3-WT CVB3-HP. Importantly, CVB3-HP showed very few adverse events. Conclusion: strong virus cancer, TNBC, due remarkable efficacy improved safety profile.

Cancer virotherapy: Targeting cancer cells by microRNA mechanism for selective replication of oncolytic viruses in these cells
Cancer, as one of the most serious public health problems, is the second-leading cause of death in the world after cardiovascular disease. The number of patients and the resulting mortality are increasing worldwide; therefore, early diagnosis, prevention, and effective treatment of cancer are very important. Current treatments such as chemotherapy and radiation therapy are often non-selective a...
متن کاملOncolytic Virotherapy in Cancer
Oncolytic viruses are natural or genetically modified viral species that selectively infect and kill neoplastic cells. Oncolytic viruses as highly targeted agents that would mediate cancer cell-autonomous anticancer effects. The therapeutic potential of oncolytic virotherapy is not a simple consequence of the cytopathic effect, but strongly relies on the induction of an endogenous immune respon...
متن کاملTargeting CXCL12/CXCR4 signaling with oncolytic virotherapy disrupts tumor vasculature and inhibits breast cancer metastases.
Oncolytic viruses hold promise for the treatment of cancer, but their interaction with the tumor microenvironment needs to be elucidated for optimal tumor cell killing. Because the CXCR4 receptor for the stromal cell-derived factor-1 (SDF-1/CXCL12) chemokine is one of the key stimuli involved in signaling interactions between tumor cells and their stromal microenvironment, we used oncolytic vir...
متن کاملOncolytic virotherapy for ovarian cancer
In the past two decades, more than 20 viruses with selective tropism for tumor cells have been developed as oncolytic viruses (OVs) for treatments of a variety of malignancies. Of these viruses, eleven have been tested in human ovarian cancer models in preclinical studies. So far, nine phase I or II clinical trials have been conducted or initiated using four different types of OVs in patients w...
