Diagnostic Performance of Direct Microscopic Techniques and Culture Media for Fungal Isolates from Hair Specimens

Objective: To compare the diagnostic performance of direct microscopic techniques and culture media for detecting fungal isolates in hair specimens.Study Design: Cross-sectional study.Place Duration Study: The study was conducted at Department Microbiology, Basic Medical Sciences Institute, Karachi, Pakistan, from January 2021 to June 2021.Materials Methods: A total 207 specimens were collected patients with superficial mycoses scalp. under aseptic using clean glass slides forceps which processed Potassium hydroxide mount Calcofluor white staining. mycological cultures performed on a Dermatophyte test medium Sabouraud Dextrose agar without antibiotics. data recorded analyzed SPSS version 21. frequencies, percentages, means, ratios calculated Descriptive statistics. association between variables by Chi-square kappa index.Results: revealed spores hyphae 154(74.3%) specimens, while staining showed elements 179(86.5%) specimens. Out 106(51.2%) positive 101(48.8%) declared negative. culture-positive dermatophytes yielded 61(57.5%) non-dermatophytes 45(42.5%) Among all media, highest number plain dextrose (59; 96.5%), followed dermatophyte (55; 90%), antibiotics (54; 90.1%). On contrary, primary isolation s 32(71.1%), 31(68.2%), 13(28.8%).Conclusion: In our study, we found technique as most sensitive method detect given For culture, tested equally effective retrieval dermatophytes. However, non-dermatophytes, SDA be better isolation.
