Differences between cultured cortical neurons by trypsin and papain digestion

چکیده
The objective of this study was to compare the efficiency trypsin and papain in neuronal digestion determine which enzyme is more efficient. Cortical tissues were obtained from Sprague–Dawley (SD) rats. According different digestive enzymes, samples divided into group group. After being digested by each two cortical neurons collected samples. Then, morphology determined. Moreover, transfected with negative control (NC) lentivirus. transfection determined compared. Compared group, number, had larger cell size, longer axonal length, fewer impurities. (57.77%) higher than that (53.83%). displayed showed body most shrank became smaller, axis mutation shorter less 6 days after NC Trypsin efficient digesting because are have a body, axons, greater efficiency.

Purified adrenocortical microsomal P-450~21 was incorporated into vesicle membranes composed of phosphatidylcholine, phosphatidylethanolamine, and phosphatidylserine a t a molar ratio of 5 3 : 1. Trypsinolysis of the incorporated P 4 5 0 ~ ~ ~ resulted in the formation of 30-, 2 5 , and 20-kDa fragments. Similar fragment formation was observed by trypsinolysis of bovine adrenocortical microsome...

The anomalies of enzyme action have frequently been accounted for by the assumption of an intermediate compound between the enzyme and the substrate. I t was shown, however, by Nelson and Hitchcock t that in the case of invertase the experiments could not be accurately calculated on this basis. The writer found with trypsin' that although the effect of varying the substrate concentration could ...
متن کاملExpression of CXCL10 in cultured cortical neurons.
Chemokines expressed in neurons are important mediators in neuron-neuron and neuron-glia signaling. One of these chemokines is CCL21 that activates microglia via the chemokine receptor CXCR3. As neurons also express CXCL10, a main ligand for CXCR3, we have thus investigated in detail the expression pattern of CXCL10 in neurons. We show that CXCL10 is constitutively expressed by neurons, is stor...

The formation of gliosis around implant electrodes for deep brain stimulation impairs electrode-tissue interaction. Unspecific growth of glial tissue around the electrodes can be hindered by altering physicochemical material properties. However, in vitro screening of neural tissue-material interaction requires an adequate cell culture system. No adequate model for cells dissociated from the inf...
