چکیده
Aberrations limit scanning fluorescence microscopy when imaging in scattering materials such as biological tissue. Model-based approaches for adaptive optics take advantage of a computational model the optical setup. Image-based adaptive optics for two-photon microscopy.
We demonstrate wavefront sensorless aberration correction in a two-photon excited fluorescence microscope. Using analysis of the image-formation process, we have developed an optimized correction scheme permitting image-quality improvement with minimal additional exposure of the sample. We show that, as a result, our correction process induces little photobleaching and significantly improves th...

Non-linear imaging is widely used in biological imaging, primarily because of its ability to image through tissue to depth of a few hundred micrometers. Because two photons need to be absorbed to excite a fluorophore in this instrument, the probability of fluorescence emission of a detectable photon scales with the intensity squared of the beam. As a result, aberrations in the beam path that re...
متن کاملAxial range of conjugate adaptive optics in two-photon microscopy.
We describe an adaptive optics technique for two-photon microscopy in which the deformable mirror used for aberration compensation is positioned in a plane conjugate to the plane of the aberration. We demonstrate in a proof-of-principle experiment that this technique yields a large field of view advantage in comparison to standard pupil-conjugate adaptive optics. Further, we show that the exten...

Two-photon fluorescence microscopy provides a powerful tool for deep tissue imaging. However, optical aberrations from illumination beam path limit imaging depth and resolution. Adaptive Optics (AO) is found to be useful to compensate for optical aberrations and improve image resolution and contrast from two-photon excitation. We have developed an AO system relying on a MEMS Deformable Mirror (...
متن کاملOptimal model-based sensorless adaptive optics for epifluorescence microscopy
We report on a universal sample-independent sensorless adaptive optics method, based on modal optimization of the second moment of the fluorescence emission from a point-like excitation. Our method employs a sample-independent precalibration, performed only once for the particular system, to establish the direct relation between the image quality and the aberration. The method is potentially ap...
