چکیده

Diosgenin (DGN), which is a sterol occurring in plants of the Dioscorea family, has attracted increasing attention for its various pharmacological activities. DGN structural similarity to cholesterol (Cho). In this study we investigated effects common tetracyclic cores and different side chains on physicochemical properties lipid bilayer membranes. Differential scanning calorimetry showed that Cho reduce phase transition enthalpy similar extent. 2H NMR, deuterated-DGN/Cho POPC ordering bilayers, revealed oriented parallel membrane normal like Cho. It was suggested affinity DGN-Cho stronger than DGN-DGN or Cho-Cho interaction. 31P NMR bilayers that, unlike Cho, altered interactions headgroups at 30 mol%. These results suggest below mol% with basic biomembrane properties.

Pairing of cholesterol with oxidized phospholipid species in lipid bilayers.

We claim that (1) cholesterol protects bilayers from disruption caused by lipid oxidation by sequestering conical shaped oxidized lipid species such as 1-palmitoyl-2-azelaoyl-sn-glycero-3-phosphocholine (PZPC) away from phospholipid, because cholesterol and the oxidized lipid have complementary shapes and (2) mixtures of cholesterol and oxidized lipids can self-assemble into bilayers much like ...

Effects of Stretching Speed on Mechanical Rupture of Phospholipid/Cholesterol Bilayers: Molecular Dynamics Simulation

Rupture of biological cell membrane under mechanical stresses is critical for cell viability. It is triggered by local rearrangements of membrane molecules. We investigated the effects of stretching speed on mechanical rupture of phospholipid/cholesterol bilayers using unsteady molecular dynamics simulations. We focused on pore formation, the trigger of rupture, in a 40 mol% cholesterol-includi...

Lateral distribution of cholesterol in dioleoylphosphatidylcholine lipid bilayers: cholesterol-phospholipid interactions at high cholesterol limit.

Lateral organization of cholesterol in dioleoyl-phosphatidylcholine (DOPC) lipid bilayers at high cholesterol concentration (>45 mol%) was investigated using steady-state fluorescence anisotropy and fluorescent resonance energy transfer techniques. The recently devised Low Temperature Trap method was used to prepare compositionally uniform cholesterol/DOPC liposomes to avoid the problem of lipi...

Condensed complexes and the calorimetry of cholesterol-phospholipid bilayers.

A recent thermodynamic model describes a reversible reaction between cholesterol (C) and phospholipid (P) to form a condensed complex C(nq)P(np). Here q and p are relatively prime integers used to define the stoichiometric composition, and n is a measure of cooperativity. The present study applies this model to the scanning calorimetry of binary mixtures of cholesterol and saturated phosphatidy...

Cholesterol-induced modulated phase in phospholipid membranes.

We report the observation of a cholesterol-induced modulated phase (Pbeta) in dipalmitoyl phosphatidylcholine bilayers. It occurs below the main transition of the lipid at cholesterol concentrations of around 15 to 20 mol % and is distinct from the ripple (Pbeta') phase found in between the main and pretransitions at lower cholesterol concentrations. An electron density map of this phase, const...
