Disulfides as redox switches: from molecular mechanisms to functional significance.

The molecular mechanisms underlying thiol-based redox control are poorly defined. Disulfide bonds between Cys residues are commonly thought to confer extra rigidity and stability to their resident protein, forming a type of proteinaceous spot weld. Redox biologists have been redefining the role of disulfides over the last 30-40 years. Disulfides are now known to form in the cytosol under condit...

Redox control of microglial function: molecular mechanisms and functional significance.

Neurodegenerative diseases are characterized by chronic microglial over-activation and oxidative stress. It is now beginning to be recognized that reactive oxygen species (ROS) produced by either microglia or the surrounding environment not only impact neurons but also modulate microglial activity. In this review, we first analyze the hallmarks of pro-inflammatory and anti-inflammatory phenotyp...

Cysteines as Redox Molecular Switches and Targets of Disease

Thiol groups can undergo numerous modifications, making cysteine a unique molecular switch. Cysteine plays structural and regulatory roles as part of proteins or glutathione, contributing to maintain redox homeostasis and regulate signaling within and amongst cells. Not surprisingly therefore, cysteines are associated with many hereditary and acquired diseases. Mutations in the primary protein ...

Molecular Rotors as Switches

The use of a functional molecular unit acting as a state variable provides an attractive alternative for the next generations of nanoscale electronics. It may help overcome the limits of conventional MOSFETd due to their potential scalability, low-cost, low variability, and highly integratable characteristics as well as the capability to exploit bottom-up self-assembly processes. This bottom-up...

Thia-bridged triarylamine heterohelicene radical cations as redox-driven molecular switches.

The first example of one-electron oxidation of thia-bridged triarylamine heterohelicenes to the corresponding exceptionally stable radical cations, fully characterized, as hexafluoroantimonate salts, by means of UV-Vis, EPR, ENDOR, density functional theory calculations and X-ray analyses, is reported. Chemical and electrochemical reversible redox processes are solidly demonstrated.
