Early detection of myocardial changes with and without dexrazoxane using serial magnetic resonance imaging in a pre-clinical mouse model

Abstract Background Cancer therapy-related cardiac dysfunction may occur in pediatric cancer survivors. Identification of early markers myocardial damage secondary to anthracycline exposure is crucial develop strategies that ameliorate this complication. Objectives The purpose study was identify changes induced by doxorubicin with and without cardioprotection using dexrazoxane detected serial magnetic resonance imaging (CMR) a pre-clinical mouse model. Methods Serial CMR examinations were performed 90 mice distributed 3 groups: 45 received (DOX group), 30 (DOX/DEX group) 15 saline injections (control group). We obtained the following parameters all mice: T2, extracellular volume quantification (ECV), deformation, functional quantification. Results Myocardial edema assessed T2 time earliest parameter demonstrating evidence injury, most notable DOX group at week 4 8 compared DOX/DEX group. Similarly, global longitudinal strain abnormal both groups. However, change persisted only ECV significantly elevated final CMR, while minimally right left ejection fraction decreased, along mass ratio time, ECV, deformation correlated ventricular volume. Conclusions identifies injury from anthracyclines. Dexrazoxne did not prevent initial edema, but inflammatory sustained. be useful for detection dysfunction. demonstrates minimizes aids recovery
