Elucidation of mRNA targets of miR-145-5p in diabetic kidney disease using bioinformatics analysis

Introduction: Diabetic kidney disease (DKD) is a major global cause of end-stage-kidney disease. In view its ongoing risk progression, the search for better biomarkers and treatment led to discovery microRNAs which regulate gene expression post-translationally. Recently, we reported trend upregulation miR-145-5p in sera type 2 diabetic patients with macroalbuminuria selected Malaysian population, concurred previous vivo vitro studies DKD. addition, miR-145 has been implicated as tumour suppressor various cancers. Methods: this study, bioinformatics tools were utilized predict mRNA targets miR-145-5p. Results: A total 683 224 experimentally-validated identified by Tarbase miRTarbase, respectively. Eighty-six (86) commonly submitted Metascape Enrichr enrichment analysis. Bioinformatics analysis literature suggested that insulin receptor substrate 1 (IRS1) was most promising target Its associated Gene Ontology terms pathways included insulin-like growth factor signalling Forkhead transcription factors (FOXO), Based on these analyses, roles IRS1 DKD proposed. Conclusion: As kidneys are heterogenous cell types mechanism miRNA cell-type-dependent, prediction particularly important DKD, improve likelihood successful experimental verification specific renal types. study attempts utilize studies, not widely done recently reported.
