چکیده
Abstract Neoantigens derived from somatic mutations in cancer cells can induce antigen‐specific T‐cell immune response for immunotherapy. However, the 3D models assessing neoepitope immunogenicity and efficacy of anti‐tumor to neoantigens are less than perfect. Here, a tumor model based on recellularized liver matrix is leveraged with HepG2 investigate T cell cytotoxic reactivity toward hepatocellular carcinoma (HCC) neoantigens. The whole exome sequencing (WES) data 364 HCC patients Cancer Genome Atlas database collected 25 highly potential immunogenic human leukocyte antigen (HLA)‐A*02:01 molecule silico predicted. Six neoantigen candidates functionally validated high by measuring cellular interferon‐ γ secretion cytotoxicity during neoantigen‐specific responses vitro. Then, minigene six identified peptides constructed minigene‐modified GFP‐HepG2 generated. Neoantigen‐specific observed secreted Granzyme B, IFN‐ , PD‐1 when targeting RLM model. Overall, provides novel strategy preclinical assessment response, which helps develop personalized vaccines immunotherapy treatments patients.

DNA methylation of tumor suppressor genes in hepatocellular carcinoma
The basic unit of chromatin is a nucleosome included an octamer of the four core histones and 147 base pairs of DNA. Posttranslational histones modifications affect chromatin structure resulting in gene expression changes. CpG islands hypermethylation within the gene promoter regions and the deacetylation of histone proteins are the most common epigenetic modifications. The aberrant patterns of...

Immunotherapy in hepatocellular carcinoma based on one or a few tumor specific antigens have shown limited antitumor efficacy. As a major suppressive factor in tumor immune response, better understanding of the role of regulatory T cells (Tregs) in hepatocellular carcinoma might be important for design of future immunotherapy-based clinical protocols. Tregs from 49 HCC patients and 40 controls ...

Black seed (Nigella sativa) is considered as a biological response modifier. Thymoquinone (TQ) is the bioactive and the most abundant constituent of the volatile oil of this seed which has been shown to possess anti-inflammatory, antioxidant and anti-neoplastic effects. In this study, the effect of TQ on HepG2 cell line was investigated in an attempt to identify its potential mechanism of actio...

متن کاملImmunodominance and functional alterations of tumor‐associated antigen‐specific CD8+ T‐cell responses in hepatocellular carcinoma
UNLABELLED Hepatocellular carcinoma (HCC) is the fifth most common malignancy worldwide with a poor prognosis and limited therapeutic options. To aid the development of novel immunological interventions, we studied the breadth, frequency, and tumor-infiltration of naturally occurring CD8(+) T-cell responses targeting several tumor-associated antigens (TAA). We used overlapping peptides spanning...
