Detection, enumeration, and sizing of planktonic bacteria by image-analyzed epifluorescence microscopy.
Epifluorescence microscopy is now being widely used to characterize planktonic procaryote populations. The tedium and subjectivity of visual enumeration and sizing have been largely alleviated by our use of an image analysis system consisting of a modified Artek 810 image analyzer and an Olympus BHT-F epifluorescence microscope. This system digitizes the video image of autofluorescing or fluoro...
متن کاملImproved technique for identification and enumeration of methanogenic bacterial colonies on roll tubes by epifluorescence microscopy.
Methanogenic fluorescent colonies can be clearly identified on roll tubes by using an epifluorescence microscope equipped with a x 2 objective. Methanogenic and nonmethanogenic colonies could be counted in roll tubes prepared from methanogenic enrichment cultures. Late-developing colonies appearing after 25 days of incubation were mainly methanogenic.
متن کاملAn improved method for counting bacteria from sediments and turbid environments by epifluorescence microscopy.
We present a new procedure for effectively detaching particle-associated bacteria by 10% (v/v) methanol and sonication which is particularly suitable for samples with a high particle load and sediments. We also optimized the sample preparation by applying the highly dsDNA-specific fluorescent stain SybrGreen I together with an optically brilliant mounting medium (polyvinylalcohol 4-88, 'moviol'...
متن کاملEpifluorescence collection in two-photon microscopy.
We present a simple model to describe epifluorescence collection in two-photon microscopy when one images in a turbid slab with an objective. Bulk and surface scattering determine the spatial and angular distributions of the outgoing fluorescence photons at the slab surface, and geometrical optics determines how efficiently the photons are collected. The collection optics are parameterized by t...
متن کاملOptimal Resting-Growth Strategies of Microbial Populations in Fluctuating Environments
Bacteria spend most of their lifetime in non-growing states which allow them to survive extended periods of stress and starvation. When environments improve, they must quickly resume growth to maximize their share of limited nutrients. Cells with higher stress resistance often survive longer stress durations at the cost of needing more time to resume growth, a strong disadvantage in competitive...
