Fluoxetine Affects Intestinal Motility via 5-HT3 and Muscarinic Receptors in ex vivo Mouse Model

چکیده

Fluoxetine, a selective serotonin reuptake inhibitor anti-depressant, causes undesirable side effects, including diarrhea and constipation. This research investigated the direct effects of fluoxetine at 0.001, 0.01, 0.1, 1, 10, 100 µM on duodenal proximal colonic tissue contractions. The investigation aimed to determine related mechanisms using an isolated mouse intestine model. Our study showed that 0.001 μM increased amplitude contraction in but decreased tissue. application higher concentrations (1, µM) reduced contractions Moreover, we found stimulatory effect tone could be prevented by pre-incubating ondansetron atropine. findings suggest inhibition was mainly mediated via 5-HT3 receptors muscarinic signaling. These might explain conflicting gastrointestinal symptoms caused fluoxetine.

5-HT3 receptors and emesis

Nausea and vomiting. Nausea and vomiting is a major problem which occurs during and after the treatment with certain drugs like anesthetics, opioidanalgesics or cytostatics. Postoperative nausea and vomiting (PONV) is called the “big” little problem. As a consequence, the choice of the anesthetic drug(s) plays a role in preventing PONV [Apfel et al., Anesthesiology, 1999; Eberhart et al., Eur J...

Serotonin Augments Gut Pacemaker Activity via 5-HT3 Receptors

Serotonin (5-hydroxytryptamine: 5-HT) affects numerous functions in the gut, such as secretion, muscle contraction, and enteric nervous activity, and therefore to clarify details of 5-HT's actions leads to good therapeutic strategies for gut functional disorders. The role of interstitial cells of Cajal (ICC), as pacemaker cells, has been recognised relatively recently. We thus investigated 5-HT...

Restraint stress stimulates colonic motility via central corticotropin-releasing factor and peripheral 5-HT3 receptors in conscious rats.

Although restraint stress accelerates colonic transit via a central corticotropin-releasing factor (CRF), the precise mechanism still remains unclear. We tested the hypothesis that restraint stress and central CRF stimulate colonic motility and transit via a vagal pathway and 5-HT(3) receptors of the proximal colon in rats. (51)Cr was injected via the catheter positioned in the proximal colon t...

The muscarinic antagonists scopolamine and atropine are competitive antagonists at 5-HT3 receptors

Scopolamine is a high affinity muscarinic antagonist that is used for the prevention of post-operative nausea and vomiting. 5-HT3 receptor antagonists are used for the same purpose and are structurally related to scopolamine. To examine whether 5-HT3 receptors are affected by scopolamine we examined the effects of this drug on the electrophysiological and ligand binding properties of 5-HT3A rec...

Luminal Cholera Toxin Alters Motility in Isolated Guinea-Pig Jejunum via a Pathway Independent of 5-HT3 Receptors

Cholera toxin (CT) is well established to produce diarrhea by producing hyperactivity of the enteric neural circuits that regulate water and electrolyte secretion. Its effects on intestinal motor patterns are less well understood. We examined the effects of luminal CT on motor activity of guinea-pig jejunum in vitro. Segments of jejunum were cannulated at either end and mounted horizontally. Th...
