Fractionation of DNA and protein from individual latent fingerprints for forensic analysis

Human touch samples represent a significant portion of forensic DNA casework. Yet, the generally low abundance genetic material combined with predominantly extracellular nature in these makes DNA-based analysis exceptionally challenging. proteins present same offer an abundant and environmentally-robust alternative. Proteogenomic methods, using protein sequence variants arising from nonsynonymous mutations, have recently been applied to may viable option looking forward. However, remains gold standard any proteomics-based methods would need consider how could be co-extracted without loss. Herein, we describe simple workflow for collection, enrichment fractionation latent fingerprint samples. This approach ensures that collected can analyzed by traditional casework while proteolytically digested via liquid chromatography-tandem mass spectrometry-based proteomics sample. Sample collection non-porous surfaces (i.e., glass) is performed through application anionic surfactant over fingermark. The sample then split into separate fractions following centrifugation enrich fraction pelleting skin cells. results indicate this permits within sample, yet highlights challenge posed trace potential degrade time. Protein deposited does not appear share limitation, robust quantities across multiple human donors. quantity quality regardless age. proteomic content consistent individual donors age, supporting future genetically variable peptide (GVP) identification.
