Expression of vascular endothelial growth factor mRNA in human preimplantation embryos derived from tripronuclear zygotes.
OBJECTIVE To detect the expression of vascular endothelial growth factor (VEGF) mRNA and/or secretion of VEGF protein by human preimplantation embryos. DESIGN Human preimplantation embryos not suitable for uterine transfer were examined for beta-actin and VEGF mRNA expression. Culture media from normally fertilized and developing preimplantation embryos were assessed for VEGF protein secretio...
متن کاملThe parental origin correlates with the karyotype of human embryos developing from tripronuclear zygotes
OBJECTIVE It has previously been suggested that embryos developing from intracytoplasmic sperm-injected (ICSI) zygotes with three pronuclei (3PN) are endowed with a mechanism for self-correction of triploidy to diploidy. 3PN are also observed in zygotes after conventional in vitro fertilization (IVF). The parental origin, however, differs between the two fertilization methods. Whereas the vast ...
متن کاملNormal birth after microsurgical enucleation of tripronuclear human zygotes: case report.
Microsurgical enucleation of a single pronucleus from each of three tripronuclear zygotes was performed and the embryos were transferred to a 38-year-old woman on day 3 after fertilization. A normal healthy baby boy was born at 38 weeks and 4 days gestation, demonstrating that with polyspermic fertilization, removal of the extra male pronucleus allows the zygote to develop normally.
متن کاملHighly efficient RNA-guided genome editing in human cells via delivery of purified Cas9 ribonucleoproteins.
RNA-guided engineered nucleases (RGENs) derived from the prokaryotic adaptive immune system known as CRISPR (clustered, regularly interspaced, short palindromic repeat)/Cas (CRISPR-associated) enable genome editing in human cell lines, animals, and plants, but are limited by off-target effects and unwanted integration of DNA segments derived from plasmids encoding Cas9 and guide RNA at both on-...
