Histidine Protonation and Conformational Switching in Diphtheria Toxin Translocation Domain

چکیده
Protonation of key histidine residues has been long implicated in the acid-mediated cellular action diphtheria toxin translocation (T-) domain, responsible for delivery catalytic domain into cell. Here, we use a combination computational (constant-pH Molecular Dynamics simulations) and experimental (NMR, circular dichroism, fluorescence spectroscopy along with X-ray crystallography) approaches to characterize initial stages conformational change happening solution wild-type T-domain H223Q/H257Q double mutant. This replacement suppresses acid-induced transition, resulting retention more stable protein structure solutions at pH 5.5 and, consequently, reduced membrane-disrupting activity. first time, report pKa values T-domain, measured by NMR-monitored titrations. Most peaks side chain spectral region are titrated pKas ranging from 6.2 6.8. However, two most up-field display little down 6, which is limiting this concentrations required NMR. These absent mutant, suggesting they belong H223 H257. The constant-pH simulations indicate that solution, range 3.0 6.5, those difficult protonate being H251 Taken together, our data demonstrate previously suggested cooperative protonation all six histidines does not occur.

pH-Triggered Conformational Switching along the Membrane Insertion Pathway of the Diphtheria Toxin T-Domain
The translocation (T)-domain plays a key role in the action of diphtheria toxin and is responsible for transferring the catalytic domain across the endosomal membrane into the cytosol in response to acidification. Deciphering the molecular mechanism of pH-dependent refolding and membrane insertion of the T-domain, which is considered to be a paradigm for cell entry of other bacterial toxins, re...
متن کاملEndosome fusion induced by diphtheria toxin translocation domain.
Endosomal cargo travels through a dynamic vesicle network en route to degradation by lysosomes or recycling through the Golgi apparatus back to the cell surface. Rab5 is a key determinant of the early endosomes by organizing effector proteins in specific subdomains and mediating early endosome fusion. We find that early endosome morphogenesis and maturation is disrupted by diphtheria toxin (DT)...
متن کاملDiphtheria Toxin and Cytosolic Translocation Factors
Diphtheria Toxin (DT) was the first investigated bacterial protein toxin. As one of the most extensively studied bacterial protein toxins, it has served as a model system for the analysis of other protein toxins (Pappenheimer, 1977). As reviewed by Pappenheimer, Loeffler identified Corynebacterium diphtheriae as the causative agent of diphtheria in 1884, and the toxin was first described in the...
متن کاملThe pH-Dependent Trigger in Diphtheria Toxin T Domain Comes with a Safety Latch.
Protein-side-chain protonation, coupled to conformational rearrangements, is one way of regulating physiological function caused by changes in protein environment. Specifically, protonation of histidine residues has been implicated in pH-dependent conformational switching in several systems, including the diphtheria toxin translocation (T) domain, which is responsible for the toxin's cellular e...
متن کاملThe role of the diphtheria toxin receptor in cytosol translocation.
The role of the receptor in the transport of diphtheria toxin (DT) to the cytosol was examined. A point-mutant form of DT, CRM 107 (CRM represents cross-reacting material), that has an 8,000-fold lower affinity for the DT receptor than native toxin was conjugated to transferrin and monoclonal antibodies specific for the cell-surface receptors T3 and Thy1. Conjugating the binding site-inactivate...
