Identification of Tumor Mutation Burden and Immune Infiltrates in Hepatocellular Carcinoma Based on Multi-Omics Analysis

We aimed to explore the tumor mutational burden (TMB) and immune infiltration in HCC investigate new biomarkers for immunotherapy. Transcriptome gene mutation data were downloaded from GDC portal, including 374 samples 50 matched normal samples. Furthermore, we divided into high low TMB groups, analyzed differential genes between them with GO, KEGG, GSEA. Cibersort was used assess cell Finally, univariate multivariate Cox regression analyses performed identify related infiltration, a risk prediction model constructed. found 10 frequently mutated genes, TP53, TTN, CTNNB1, MUC16, ALB, PCLO, MUC, APOB, RYR2, ABCA. Pathway analysis indicated that these TMB-related mainly enriched PI3K-AKT. showed memory B cells ( p = 0.02), CD8+ T 0.09), CD4+ activated 0.07), neutrophils 0.06) demonstrated difference groups. On analysis, GABRA3 0.05), CECR7 < 0.001), TRIM16 0.003), IL7R 0.04) associated infiltration. The had an area under curve (AUC) of 0.69, suggesting patients better survival outcomes. Our study first time CECR7, GABRA3, IL7R, TRIM16L promoted antitumor immunity HCC.
