Identification of whitefly (Bemisia tabaci) proteins interacting with Tomato leaf curl Bangalore virus coat protein gene using Y2H system

چکیده
Tomato leaf curl Bangalore virus (ToLCBV) (Geminiviridae) causes the economically important tomato disease (ToLCVD), and is transmitted by whitefly, Bemisia tabaci (Gennadius) (Aleyrodidae: Hemiptera). Successful transmission of insect requires safe translocation through different barriers inside vector. During translocation, viral coat protein will interact with many whitefly proteins, while a few them are beneficial to (e.g. heat shock proteins) but some may be harmful immune proteins). We carried out yeast two hybrid (Y2H) assays identify proteins B.tabaci Asia 1 genetic group interacting ToLCBV (CP). The Y2H assay initially identified total 425 putative on low stringent selection media, they were reduced 324 when colonies grown high which about 274 produced single bands in colony PCR experiments remaining multiple bands. Further, pressure confirmed 102 CP these included (HSPs) 70 kDa, GroEL, nucleoproteins, vitellogenins, apolipophorins, 40 s ribosomal sorbitol dehydrogenase, dipeptidyl peptidase, E3 ubiquitin, annexin, GTP cyclohydrolase, tropomyosin, salivary secreted succinate lachesins, enolase others. could potential targets for novel control strategies such as using RNAi or insecticide target sites developing future strategies.

Replication of Tomato Yellow Leaf Curl Virus in Its Whitefly Vector, Bemisia tabaci.
UNLABELLED Tomato yellow leaf curl virus (TYLCV) is a begomovirus transmitted exclusively by the whitefly Bemisia tabaci in a persistent, circulative manner. Replication of TYLCV in its vector remains controversial, and thus far, the virus has been considered to be nonpropagative. Following 8 h of acquisition on TYLCV-infected tomato plants or purified virions and then transfer to non-TYLCV-hos...

Tomato yellow leaf curl virus (TYLCV) is a single-stranded (ssDNA) begomoviruses that causes severe damage to tomato and several other crops worldwide. TYLCV is exclusively transmitted by the sweetpotato whitefly, Bemisia tabaci in a persistent circulative and propagative manner. Previous studies have shown that the transmission, retention, and circulation of TYLCV in its vector involves intera...
متن کاملThe circulative pathway of begomoviruses in the whitefly vector Bemisia tabaci - insights from studies with Tomato yellow leaf curl virus
Our current knowledge concerning the transmission of begomoviruses by the whitefly vector Bemisia tabaci is based mainly on research performed on the Tomato yellow leaf curl virus (TYLCV) complex and on a number of viruses originating from the Old World, such as Tomato leaf curl virus, and from the New World, including Abutilon mosaic virus, Tomato mottle virus, and Squash leaf curl virus. In t...

متن کاملRate of Tomato yellow leaf curl virus Translocation in the Circulative Transmission Pathway of its Vector, the Whitefly Bemisia tabaci.
ABSTRACT Whiteflies (Bemisia tabaci, biotype B) were able to transmit Tomato yellow leaf curl virus (TYLCV) 8 h after they were caged with infected tomato plants. The spread of TYLCV during this latent period was followed in organs thought to be involved in the translocation of the virus in B. tabaci. After increasing acquisition access periods (AAPs) on infected tomato plants, the stylets, the...
