In Situ Monitoring of Drug Precipitation from Digesting Lipid Formulations Using Low-Frequency Raman Scattering Spectroscopy

Low-frequency Raman spectroscopy (LFRS) is a valuable tool to detect the solid state of amorphous and crystalline drugs in dosage forms transformation between different polymorphic forms. It has also been applied track solubilisation as suspensions milk infant formula during vitro digestion. This study reports use LFRS an approach probe drug precipitation from lipid-based delivery system (medium-chain self-nanoemulsifying system, MC-SNEDDS) Upon lipolysis digestible components MC-SNEDDS containing fenofibrate model drug, sharp phonon peaks appeared at low-frequency spectral region (<200 cm−1), indicating form formulation. Two multivariate data analysis approaches (principal component partial least squares discriminant analysis) one univariate (band ratios) were explored these changes over time. The produces results good agreement with situ small angle X-ray scattering (SAXS) measurements all used, whereas mid-frequency requires PLS-DA gain similar results. suggests that can be used complementary, potentially more accessible, technique SAXS determine kinetics formulations.
