Inverse associations between cerebellar inhibition and motor impairment in spinocerebellar ataxia type 3

چکیده

BackgroundCerebellar ataxia generally results from a lesion disrupting the corticopontocerebellar or cerebellothalamocortical tract. The cerebellar inhibition (CBI) paradigm represents dual-coil transcranial magnetic stimulation protocol that interrogates integrity of latter pathway. Whether CBI has clinical relevance in patients remains largely unknown because associations with pertinent disease severity measures etiologically homogeneous cohorts have not been previously examined.ObjectiveTo investigate if correlates and functional indices individuals spinocerebellar type 3 (SCA3).MethodsCBI was assessed fourteen SCA3 by paired-pulse cerebellar-motor cortex (M1) using interstimulus intervals 3, 5, 10 ms. Correlation coefficients were determined between severity, manual dexterity, walking speed.ResultsSuppression M1 excitability occurred 5 ms following contralateral conditioning stimulus patients, but, on average, significantly reduced as compared to healthy control group literature (p < 0.001). A significant association found decreased levels higher Scale for Assessment Rating Ataxia (SARA) scores (r = −0.62, p 0.019). negatively correlated axial, appendicular, speech subscores, well nine-hole peg test performance −0.69, 0.006). No observed speed. As expected, there no clinical-neurophysiological correlations at intervals.ConclusionOur provide first neurophysiological evidence an inverse tract integrity, reflected CBI, patients. Longitudinal studies are required evaluate could serve marker progression.

Early changes in cerebellar physiology accompany motor dysfunction in the polyglutamine disease spinocerebellar ataxia type 3.

The relationship between cerebellar dysfunction, motor symptoms, and neuronal loss in the inherited ataxias, including the polyglutamine disease spinocerebellar ataxia type 3 (SCA3), remains poorly understood. We demonstrate that before neurodegeneration, Purkinje neurons in a mouse model of SCA3 exhibit increased intrinsic excitability resulting in depolarization block and the loss of the abil...

Cognitive impairment in spinocerebellar ataxia type 8.

OBJECTIVES Only a limited number of studies have investigated the cognitive performances of spinocerebellar ataxia (SCA) patients. In none of the SCA8 studies have the neuropsychological test performances been the primary measures. The objective of the current study was to investigate the characteristics of cognitive deficits in SCA8. METHODS Ten SCA8 patients and ten case-by-case matched con...

Cognitive impairment in Spinocerebellar ataxia type 10

Background: Cognitive and psychiatric dysfunction has been described in several neurodegenerative diseases but has not been systematically evaluated in spinocerebellar ataxia type 10 (SCA10). Objective: The aim of the present study was to investigate the core cognitive features in a large cohort of Brazilian patients with SCA10, comparing the results against a healthy control group. Methods: Tw...

Spinocerebellar ataxia type 6 protein aggregates cause deficits in motor learning and cerebellar plasticity.

Spinocerebellar ataxia type 6 (SCA6) is linked to poly-glutamine (polyQ) within the C terminus (CT) of the pore-forming subunits of P/Q-type Ca(2+) channels (Cav2.1) and is characterized by CT protein aggregates found in cerebellar Purkinje cells (PCs). One hypothesis regarding SCA6 disease is that a CT fragment of the Cav2.1 channel, which is detected specifically in cytosolic and nuclear frac...

Cerebellar Soluble Mutant Ataxin-3 Level Decreases during Disease Progression in Spinocerebellar Ataxia Type 3 Mice

Spinocerebellar Ataxia Type 3 (SCA3), also known as Machado-Joseph disease, is an autosomal dominantly inherited neurodegenerative disease caused by an expanded polyglutamine stretch in the ataxin-3 protein. A pathological hallmark of the disease is cerebellar and brainstem atrophy, which correlates with the formation of intranuclear aggregates in a specific subset of neurons. Several studies h...
