Macrophage Polarization as a Facile Strategy to Enhance Efficacy of Macrophage Membrane‐Coated Nanoparticles in Osteoarthritis

چکیده
Osteoarthritis (OA) is a chronic degenerative joint disorder associated with pain and inflammation, the leading cause of disability worldwide. Owing to complexity OA inflammation driven by plethora inflammatory cytokines, current specific anti-cytokine therapies have not been successful. Among immune cells implicated in macrophages reportedly regulate via macrophage polarization. Given that pro-inflammatory M1 anti-inflammatory M2 opposing roles exploiting advanced polarization subsets (M0, M1, M2) enhance therapeutic efficacy membrane-coated gold (Au) nanoparticles (NPs) as broad-spectrum agent for treatment proposed. Herein, it shown among NPs generated from various subsets, (Au-M2 NPs) uniquely exhibit superior sponging cytokines alleviating matrix degradation over its counterparts derived same cell source, both inflammation-stimulated chondrocyte explant models. Collectively, herein described results validate facile strategy membrane NP-based immunotherapy potential treatment.

Macrophage/microglia with heterogonous phenotype and function under physiological and pathological conditions are the main cell lineage involved in inducing immune responses in neuroinflammatory diseases which exhibit combined inflammatory and anti-inflammatory functions. An increase in the expression of iNOS triggers M1 phenotype that secrete high concentrations of inflammatory cytokines, whil...

Hypoxia in ischemic limbs typically initiates angiogenic and inflammatory factors to promote angiogenesis in attempt to restore perfusion, and revascularization involves multiple cell types and systems. Macrophage display phenotype plasticity, and can polarize in response to local and systemic cytokine stimuli. M2 macrophage are known to play an important role in angiogenesis and wound healing....

متن کاملMacrophage Polarization-Introduction
Macrophages (MΦ) are phagocytic cells of the innate immune system that are present in virtually all tissues. Macrophages differentiate from circulating peripheral-blood mononuclear cells (PBMCs), which migrate from the bloodstream into tissue in the steady state or in response to trauma or infection [1]. Mononuclear cells develop from myeloid progenitor cells in the bone marrow that serve as th...

In the 1990s it was discovered that the cytokine interleukin (IL)-4 induced different effects on macrophage gene expression compared to that of interferon (IFN)-gamma and lipopolysaccharide (LPS). In contrast to the classical activation of macrophages by IFN-gamma (Nathan CF et al. 1983), the macrophage gene expression induced by IL-4 was described as “alternative activation” (Stein M et al. 19...
