چکیده

Introduction Long non-coding RNA Membrane-Associated Guanylate Kinase Inverted 2 Antisense 3 (MAGI2-AS3) has been identified as a predictive characteristic for prostate cancer (PC). However, the underlying mechanism of MAGI2-AS3 in PC is yet unclear. Material and methods The gene expression was detected via reverse transcription quantitative PCR. Cell viability, apoptosis invasion were cell counting kit-8, caspase-3 activity, flow cytometry, transwell assays. Tumor xenograft experiment performed to measure tumor growth vivo. Luciferase radioimmunoprecipitation assays utilized determine association between microRNA (miR)-142-3p or proline-rich membrane anchor 1 (PRIMA1). Results PRIMA1 established be downregulated PC, whereas miR-142-3p exhibited upregulation PC. In vitro vivo loss-of-function demonstrated that silencing increased viability invasiveness, enhanced cells, while reducing apoptosis. cells showed similar effect silencing. Moreover, inhibitor reversed impacts downregulation on malignant behavior cells. Conclusions knockdown by targeting suppress expression. Our findings reveal can promising therapeutic target treatment

Membrane-associated guanylate kinase with an inverted arrangement of protein-protein interaction domains (MAGI)2 (also called synaptic scaffolding molecule (S-SCAM), atrophin-1-interacting protein 1, activin receptor-interacting protein 1) is a scaffold protein that binds a wide variety of receptors, cell adhesion molecules and signalling molecules. It also interacts with other scaffold protein...

miR-142-3p Contributes to Early Cardiac Fate Decision of Embryonic Stem Cells

MicroRNAs (miRNAs) play important roles in cell fate decisions. However, the miRNAs and their targets involved in the regulation of cardiac lineage specification are largely unexplored. Here, we report novel functions of miR-142-3p in the regulation of cardiomyocyte differentiation from mouse embryonic stem cells (mESCs). With a miRNA array screen, we identified a number of miRNAs significantly...

بررسی تغییرات بیانmiR-101و ارتباط آن با میزان بیان ژن‌های سرکوب‌کننده تومور PTENو MAGI2در سه رده سلولی سرطان پروستات

Introduction: Defects in PTEN gene play an important role in the initiation of the prostate cancer. MAGI2 is recognized as a frame which assists in stabilizing PTEN protein and enhances its tumor suppressor function. As miR-101 down-regulates MAGI2 expression, it can indirectly cause to reduce the PTEN activity. Methods:LnCap, PC3 and DU-145 prostate cancer cell lines were studied in order to ...

MiR-487a Promotes TGF-β1-induced EMT, the Migration and Invasion of Breast Cancer Cells by Directly Targeting MAGI2

Tumor metastasis is a complex and multistep process and its exact molecular mechanisms remain unclear. We attempted to find novel microRNAs (miRNAs) contributing to the migration and invasion of breast cancer cells. In this study, we found that the expression of miR-487a was higher in MDA-MB-231breast cancer cells with high metastasis ability than MCF-7 breast cancer cells with low metastasis a...

miR‑142‑3p promotes osteoblast differentiation by modulating Wnt signaling.

Canonical Wnt signaling is critical for the control of osteoblast differentiation in human mesenchymal stem cells. MicroRNAs (miRs) are essential regulators of cell differentiation by post‑transcriptional regulation of target gene expression. The aim of the present study was to investigate the molecular mechanism by which miR‑142‑3p promotes osteoblastic differentiation using the human fetal os...
