Direct measurement of the mechanical work during translocation by the ribosome
A detailed understanding of tRNA/mRNA translocation requires measurement of the forces generated by the ribosome during this movement. Such measurements have so far remained elusive and, thus, little is known about the relation between force and translocation and how this reflects on its mechanism and regulation. Here, we address these questions using optical tweezers to follow translation by i...
متن کاملMovement in ribosome translocation
Translocation of peptidyl-tRNA and mRNA within the ribosome during protein synthesis is promoted by the elongation factor EF-G and by the hydrolysis of GTP. A new study reports that EF-G binds to ribosomes as an EF-G.GDP complex and that GTP is exchanged for GDP on the ribosome. Together with cryo-electron microscopy, this unexpected finding helps clarify the role of GTP in translocation.
متن کاملStructural insights into ribosome translocation
During protein synthesis, tRNA and mRNA are translocated from the A to P to E sites of the ribosome thus enabling the ribosome to translate one codon of mRNA after the other. Ribosome translocation along mRNA is induced by the universally conserved ribosome GTPase, elongation factor G (EF-G) in bacteria and elongation factor 2 (EF-2) in eukaryotes. Recent structural and single-molecule studies ...

Small GTPases play central roles in catalyzing each stage of protein synthesis on the ribosome. In prokaryotes, the relevant GTPases are: initiation factor IF2, which delivers the initiator tRNA to the P (peptide) site of the 30S ribosomal subunit; elongation factor EF-Tu, which delivers the aminoacyl-tRNA to the 70S ribosome (composed of 50S and 30S subunits); elongation factor EF-G, which pro...
متن کاملStructural insights into Pre-Translocation Ribosome Motions
Subsequent to the peptidyl transfer step of the translation elongation cycle, the initially formed pre-translocation ribosome, which we refer to here as R(1), undergoes a ratchet-like intersubunit rotation in order to sample a rotated conformation, referred to here as R(F), that is an obligatory intermediate in the translocation of tRNAs and mRNA through the ribosome during the translocation st...
