Automated synthesis of backbone protected peptides†
Solid phase peptide synthesis (SPPS) is a powerful technology for the chemical synthesis of peptides and small proteins. However, access to many targets is often complicated and sometimes precluded by the occurrence of so-called difficult sequences. When encountered during SPPS difficult sequences are associated with a collapse of the swollen resin volume, incomplete acylation and in the case o...
متن کاملOne-step labelling of oligonucleotides with fluoresceine during automated synthesis.
Fluorescent or biotin labelling of an oligonucleotide at it's 5'-end usually involves two steps. First, a N-protected aminoalkyl phosphoramidite derivative (as for instance Aminolink 2 from Applied Biosystems) is added to the 5'-end of an oligonucleotide during automated DNA synthesis. After removal of all protecting groups the NHS ester of an appropriate fluorescent dye or biotin is coupled to...
متن کاملOligonucleotides with novel, cationic backbone substituents: aminoethylphosphonates.
Oligonucleotide (2-aminoethyl)phosphonates in which the backbone consisted of isomerically pure, alternating (2-aminoethyl)-phosphonate and phosphodiester linkages have been prepared and characterized. One of these single isomer oligonucleotides (Rp) formed a more stable duplex with DNA or RNA than its corresponding natural counterpart. Hybrid stability was more pH-dependent, but less salt-depe...
متن کاملSynthesis and properties of DNA oligonucleotides with a zwitterionic backbone structure.
The nucleosyl amino acid (NAA)-modification of oligonucleotides is introduced, which enables the preparation of oligonucleotides with zwitterionic backbone structures. It is demonstrated that partially zwitterionic NAA-modified DNA oligonucleotides are capable of duplex formation with native polyanionic counterstrands and show retained sensitivity towards base-pairing mismatches.
متن کاملDual polarisation interferometry for real-time, label-free detection of interaction of mercury(II) with mercury-specific oligonucleotides.
A real-time, label-free dual polarisation interferometry technique was used to investigate the interaction of Hg(2+) with a 21-mer T-rich oligonucleotide and further construct a Hg(2+) biosensor based on thymine-Hg(2+)-thymine coordination chemistry.
