"“Mitochondrial Cells” Can Live Without a Nucleus, and Possess Proteins Encoded by Both Nuclear and Mitochondrial DNA"

چکیده

Nakano first discovered the “Mitochondrial cells” (Mito Cells), which can survive for long term despite lacking a nucleus. However, there was no clear evidence to indicate whether MitoCells were alive. To resolve this question, we constructed GFP-actin induced Mito Cell (GFP-actin Cell). Herein, report molecular analysis of both and MitoCells. Our results suggest that have minimum amounts proteins DNA needed sustain life.

Mammalian sperm translate nuclear-encoded proteins by mitochondrial-type ribosomes.

It is widely accepted that spermatozoa are translationally silent. The present study demonstrates, for the first time, incorporation of labeled amino acids into polypeptides during sperm capacitation, which was completely inhibited by mitochondrial translation inhibitors but not by the cytoplasmic translation inhibitor. Unlike 80S cytoplasmic ribosomes, 55S mitochondrial ribosomes were present ...

Mitochondrial heat shock protein 70, a molecular chaperone for proteins encoded by mitochondrial DNA

Mitochondrial heat shock protein 70 (mt-Hsp70) has been shown to play an important role in facilitating import into, as well as folding and assembly of nuclear-encoded proteins in the mitochondrial matrix. Here, we describe a role for mt-Hsp70 in chaperoning proteins encoded by mitochondrial DNA and synthesized within mitochondria. The availability of mt-Hsp70 function influences the pattern of...

CLUH regulates mitochondrial biogenesis by binding mRNAs of nuclear-encoded mitochondrial proteins

Mitochondrial function requires coordination of two genomes for protein biogenesis, efficient quality control mechanisms, and appropriate distribution of the organelles within the cell. How these mechanisms are integrated is currently not understood. Loss of the Clu1/CluA homologue (CLUH) gene led to clustering of the mitochondrial network by an unknown mechanism. We find that CLUH is coregulat...

Mitochondrial DNA Mutations, Pathogenicity and Inheritance

Mitochondria contain their own DNA (mtDNA), which codes for 13 proteins (all subunits of the respiratory chain complexes), 22 tRNAs and 2 rRNAs. Several mtDNA point mutations as well as deletions have been shown to be causative in well-defined mitochondrial disorders. A mixture of mutated and wild type mtDNA (heteroplasmy) is found in most of these disorders. Inheritance of mtDNA is maternal, a...

A REPORT OF RAPID AND EFFICIENT METHOD FOR MITOCHONDRIAL DNA

Based on the similarity of prokaryotic DNA to that of mitochondrial DNA, a rapid and efficient method for DNA purification from mitochondrial extracts obtained from different sources, was carried out based on the modification method described by Marmur (1983) which resulted in a high degree of purity (A260/A280=2). In addition we have applied the same technique for the first time to purify...
