Morpho-molecular and Pathogenic Variability of Wilt of Lentil from Indo-Gangetic Plains of India

Background: Fusarium wilt in lentil (Lens culinaris L.) infected by oxysporum is considered as one of the major biotic factors for low productivity lentil. The present study was conducted to determine nine isolates (W1-W8 and W10) which were isolated from different growing fields West Bengal, India characterized their physio-morphological, biochemical molecular traits vitro. Methods: Nine plant samples collected 9 places Bengal. Pathogen using PDA media followed cultural, morphological, pathogenic variability studies all Fusarium. Type pigmentation, sporulation, conidia size each isolate recorded observing culture plate after complete growth mycelium. Pathogenicity test done check aggressiveness three varieties (Maitri, HUL-57 BM-6). fungal DNA amplified with specific primer pair, Forward ITS 1 Reverse 4. Result: Morphological assessment showed distinct identification characters variability. mycelia septate, profusely branched differential texture margin.All produced micro-and macro-conidia pure within seven days inoculation morphological pathogenicity test, variety Maitri better germination (100% W2, W4, W6, W7, W8 less disease severity against most strains (75% W1) compared other two hence taken into consideration greenhouse field trials. Furthermore, performed sequence analysis rDNA-ITS region sequencing supported confirmed associated fungi at species level.
